Abstract:
Disclosed in the present application is a method of detecting a back mutation in a virus sample. The method comprises: obtaining a nucleic acid in the virus sample; obtaining a reaction mixture of polymerase chain reaction (PCR) containing the nucleic acid, a primer, a nucleotide monomer mixture and a nucleic acid polymerase, wherein the primer can specifically bind to the back-mutated nucleic acid via complementary base pairing; dispersing the PCR reaction mixture into a plurality of micro-droplets; separately performing PCR amplification on the plurality of micro-droplets; detecting the PCR amplification product in the plurality of micro-droplets. Further disclosed in the present application is a kit for implementing the method.
