Abstract:
Disclosed is a control detection method which can detect both a positive control and a negative control simultaneously and conveniently in one and the same reaction system. A template nucleic acid for a control is added to a reaction system in which the control is to be detected, and an amplification reaction is subsequently carried out. The template nucleic acid can be amplified with a primer that enables the amplification of a target sequence of interest. A detection probe capable of hybridizing with the target sequence can hybridize with an amplification region that is contained in the template nucleic acid for the control and can be amplified with the primer. The Tm value (TmC) between the amplification region that is contained in the template nucleic acid for the control and can be amplified with the primer and the detection probe is so set as to become different from a Tm value (TmA) between the target sequence and the detection probe. The occurrence of amplification in the reaction system can be determined when a peak appears at the TmC, and the occurrence of the contamination of the reaction system by any undesired nucleic acid can be determined when a peak appears at a temperature other than the TmC.
