Abstract:
The present invention provides a method for detecting the presence or absence of guanine-abasic sites, the method being a process for detecting guanines opposite abasic sites that occur in double stranded DNA. The method comprises: (1) a first step for position-selectively cutting an abasic site in double stranded DNA using oxygen; (2) a second step for modifying the 2-position amino group of a guanine that is opposite to an abasic site using a modifier; and (3) a third step for performing a polymerase chain reaction having the modified double stranded DNA obtained by performing the first and second steps as a template and checking for the presence or absence of amplification products. (The order of the first step and the second step does not matter.)
