Abstract:
Provided is a method for detecting intercellular interaction. The method comprises: (1) preparing a first cell expressing a TCR activation response element and a reporter molecule, the first cell expressing a TCR; (2) co-incubating an antigen peptide with a second cell expressing an MHC to give a second cell loaded with the antigen peptide; (3) marking the first cell using a first label to give a first cell carrying the label, and marking the second cell loaded with the antigen peptide using a second label to give a second cell carrying the label; (4) encapsulating the first cell carrying the label, the second cell carrying the label, and a magnetic bead using an oil phase to prepare a plurality of liquid droplets; (5) injecting a cell lysis buffer into a single liquid droplet, and sequencing nucleic acids after lysis; and (6) determining, according to the sequencing result, whether the first cell and the second cell in the single liquid droplet have interaction with each other.
