Abstract:
The present invention relates to a method for isolating and/or detecting apolipoprotein from a biological sample by preprocessing with an organic solvent. Additionally, the invention relates to a matrix and internal standard composition for mass analysis of proteins including sera from ruminant animals. The present invention not only allows direct quantification of various proteoforms even without enzymatic cleavage, but also uses animal serum as a substrate to simply maintain the matrix of the sample without removing the target protein in the sample, and the objectivity and accuracy of quantitative results were ensured at minimal cost by using proteins in animal serum as internal standards instead of existing synthetic peptides or synthetic proteins that requires time and cost for their synthesis. Thus, the present invention can be advantageously used as an efficient analytical method that enables the extraction of target proteins with high purity from samples and provides highly reliable quantitative information on individual proteoforms with various glycosylation patterns, as well as information on the total amount of all proteoforms in the sample.
