Abstract:
The invention provides a safe and efficient method for in vitro transduction of double-negative T cells (DNT), and cells prepared using the method, and a preparation containing the cells. In the present invention, under the condition that a transduction-assisting cationic reagent is not added, stable and efficient transduction efficiency can be obtained using fresh enriched or cryopreserved recovered double-negative T cells which are activated and amplified by lentivirus transduction. A chimeric antigen receptor - double-negative T cell (CAR-DNT) prepared using the method of the present invention not only has higher cell viability and purity, but also maintains high tumor-killing activity, and avoids the high residual hidden dangers caused by adding a cationic transduction reagent during CAR-DNT cell production.
