Abstract:
Disclosed is a method for highly sensitive biomarker quantification using photo-oxidation amplification. The method for highly sensitive biomarker quantification using photo-oxidation amplification according to an embodiment of the present invention comprises the steps: of performing an enzyme-substrate reaction on a measurement sample containing an enzyme labeled on any one selected from an antibody, an aptamer and a nucleic acid that specifically bind to a biomarker; performing a photo-oxidation amplification process by continuously exposing light to a product of the enzyme-substrate reaction, while optically measuring any one or more optical characteristics selected from a color development amount, a luminescence amount and a fluorescence amount of a product during the photo-oxidation amplification process; indexing changes over time with respect to the measured optical characteristics; and comparing indices extracted from the indexing step with the index of a reference sample so as to quantify the concentration of the biomarker contained in the measurement sample. Therefore, the method of the present invention has an effect of more accurately and highly sensitively quantifying a low concentration or a trace of biomarker, by continuously exposing light to a product of a reaction of a substrate with an enzyme labeled on an antibody, an aptamer, a nucleic acid, etc. specific to a biomarker to be detected so as to perform a photo-oxidation amplification process, and indexing changes in the optical characteristics of the product over time.
