Abstract:
 PCR accuracy is critical for PCR amplification when preparing a library for cloning or sequential analysis. Therefore, there is a demand for increased accuracy, even for family B DNA polymerases that are said to be highly accurate. In particular, even family B DNA ribosomes that are said to be highly accurate are susceptible to repeat array errors, and there is a demand for a reduction of repeat array errors. The objectives of the present invention are to provide a method for improving the accuracy of nucleic acid amplification, and to provide a method for increase the accuracy of nucleic acid amplification by reducing repeat array errors. By adding PCNA to family B DNA ribosomes, it was possible to increase the accuracy of nucleic acid amplification. Moreover, the addition of PCNA made it possible to reduce repeat array errors and increase the accuracy of nucleic acid amplification.
