Abstract:
Disclosed is a method for increasing the mass spectrum identification number of a protein and/or peptide fragment group. The method comprises: step S1, adding a binding buffer solution to a sample under test, and adding an aluminosilicate molecular sieve to obtain a suspension; step S2, after incubating the suspension, performing high-speed centrifugation, removing the supernatant, and retaining a precipitate I; step S3, re-suspending the precipitate I by using a cleaning buffer solution to obtain a re-suspension, centrifuging same at a high speed, then removing the supernatant, retaining a precipitate II, and repeating step S3 several times; and step S4, preparing the precipitate II into a protein group sample and performing mass spectrum detection. By means of the aluminosilicate molecular sieve and the corresponding binding buffer solution, different types of proteins and peptide fragments can be effectively adsorbed, and the selective enrichment of low-abundance proteins and peptide fragments is realized. The interference of high-abundance proteins on the identification of low-abundance proteins during mass spectrum identification is effectively avoided. The method can be used for identifying various protein spectra, and the processes of trace sample enrichment, enzymatic desalting, etc. at the protein level or peptide fragment level can be performed.
