Abstract:
Disclosed in the present invention are a method for inducing the differentiation of iPSCs to obtain CD34+ cells and NK cells and the use thereof. According to the method, using the three-dimensional structure of an embryoid body to provide a good differentiation microenvironment for the differentiation of iPSC, a high proportion of CD34+ cells can be produced on day 4 under hypoxia-induced culture conditions, and subsequently, by means of inducing the differentiation via embryoid body adhesion or digestion and resuspension, the yield of NK cells obtained by the induced differentiation of iPSCs is significantly improved, and the NK cells obtained by means of the induced differentiation can play a role in killing tumor cells in a short time, and have strong tumor-killing ability. The method is suitable for the large-scale production and clinical application of cell preparations.
