Abstract:
The purpose of the present invention is to provide a method for inducing the differentiation of human pluripotent stem cells into human primordial germ cell-like (PGC-like) cells at a high efficiency and high reproductivity, and a cell surface marker for specifying human PGC-like cells. To achieve the above purpose, provided is a method for producing human PGC-like cells from human pluripotent stem cells, said method comprising: (I) a step for culturing human pluripotent stem cells in a liquid culture medium containing activin A and a GSK3β inhibitor to give mesoderm-like cells; and (II) a step for culturing the mesoderm-like cells obtained in step (I) in a liquid culture medium containing BMP. Also provided is a method for producing isolated human PGC-like cells, said method comprising: (III) a step for, among the cells obtained in the aforesaid step (II), screening cells positive for at least one cell surface maker selected from the group consisting of PECAM (CD31), INTEGRINα6 (CD49f), INTEGRINβ3 (CD61), KIT (CD117), EpCAM, PODOPLANIN and TRA1-81.
