Abstract:
Method of labeling immunoglobulin using quantum dots comprises steps of cleaning quantum dots, performing activation, cleaning the quantum dots a second time, cleaning immunoglobulin, performing coupling, and performing separation. By means of structural characteristics of water-soluble quantum dots, characteristics of reactions of activation of carboxyl groups with EDC and characteristics of a condensation reaction, and through optimization of a condition of each step, carboxyl groups on surfaces of quantum dots are completely activated and are in a suitable labeling environment. Through the optimization of a condition of a coupling step, all quantum dots can be bonded to the surface of immunoglobulin through a condensation reaction, so that labeling efficiency is high. By using a reaction solution obtained after steps of exclusion, chromatography, separation, purification, and coupling, immunoglobulin that has not undergone reaction can be recycled for continued labeling, thereby improving utilization of immunoglobulin and saving cost. A manufactured protein-quantum dot conjugate has high activity and simple operations and used devices, and can achieve industrial scaled-up production.
