Abstract:
Disclosed are: a method for increasing the change in voltage or ionic strength of a voltage-sensitive fluorescent dye; and the measurement of the change in action potential of a cardiomuscular cell derived from an ES cell or an iPS cell, which is never before impossible to achieve. The screening of various substances is carried out. As a result, it is found that vitamin E has an activity of increasing the sensitivity of a voltage-sensitive fluorescent dye and cholesterol has an activity of increasing the fluorescence intensity of a voltage-sensitive fluorescent dye. It is also found that these substances can be used in combination to increase the sensitivity of a voltage-sensitive fluorescent dye with vitamin E while increasing the absolute fluorescence intensity of the voltage-sensitive fluorescent dye with cholesterol.
