Abstract:
A method for measuring the optical purity of an ezetimibe intermediate, characterized in that: a chromatographic column for which amylose-tris(3-chloro-4-methylphenylcarbamate) is bonded on the surface of silica gel as a filler is employed, and n-hexane-ethanol-dichloromethane serves as mobile phase. The measuring method is such that: preparing an ezetimibe intermediate sample and a racemic solution at 0.46-0.54 mg/ml; extracting 20 μl of the ezetimibe intermediate racemic solution and injecting same into a liquid-phase chromatograph, when the degree of separation between two isomer peaks is greater than 1.5, extracting 20 μl of an ezetimibe sample and injecting same into the liquid-phase chromatograph, and calculating the optical purity by means of a peak area normalization method. The method of the present invention for measuring the optical purity of the ezetimibe intermediate uses a short amount of time, where R > 1.5. Also, with great peak shapes being provided, the optical purity of the ezetimibe intermediate can be accurately measured, thus controlling the quality of an ezetimibe final product.
