Abstract:
The present invention belongs to the technical field of the preparation of flexible hybrid film materials and probes for visual detection, and particularly relates to a method for preparing an up-conversion luminescence flexible hybrid film for visual detection of tumor markers. Metal-ion-doped black phosphorus quantum dots M-BPQDs are prepared by using ultrasonic and solvothermal processes; mesoporous SiO2 grows on the surfaces thereof and amination modification is performed; the M-BPQDs are connected to a carboxylated single-stranded DNA1; receptor molecules enter holes; single-stranded DNA2 aptamers and the DNA1 are bonded due to base complementation to package receptors in the holes; and an M-BPQD probe is prepared. DNA1 terminal-SH and a composite film formed by assembling polymethyl methacrylate-polyimide-gold nanoparticles in a layer-by-layer manner are bonded by Au-S bonds, and the film and the probe are connected to construct a flexible hybrid film. Compared with the prior art, the flexible hybrid film of the present invention is simple to prepare, low cost and highly sensitive; when a biological fluid sample containing a tumor marker is added in a dropwise manner to the hybrid film, a relationship between the up-conversion luminescence intensity and the concentration of the marker is constructed under the excitation of near-infrared light, so that the visual detection of the marker is realized.
