Claims:
WHAT CLAIMED IS

1. A non-therapeutic method for preventing, mitigating or treating PTGS2-induced disorders of human skin and related dysfunctions, comprising or consisting of the following step: applying a working amount of a carnosine compound according to formula (I) in which R1 stands for H or CH3 and R2 for H or COOH, to human skin

2. The method of Claim 1, wherein the skin disorder is inflammation, ageing and/or skin cancer.

3. The method of Claim 1, wherein the dysfunction is fever and/or pain.

4. The method of Claim 1, wherein said carnosine compound is selected from the group consisting of Carnosine, L-Carnosine, D-Carnosine, D/L-Carnosine, Anserine, D- Anserine, L-Anserine, L-Anserine*HNO3 and mixtures thereof.

5. The method of Claim 1, wherein said carnosine compound is L-Carnosine.

6. The method of Claim 1, wherein said carnosine compound is applied to human skin as a component of a cosmetic skin or sun care formulation.

7. The method of Claim 6, wherein said cosmetic formulation is a cream, a lotion, an ointment or a spray.

8. The method of Claim 6, wherein said carnosine compounds are present in said cosmetic formulations in amounts of from about 0.01 to about 5 wt.-percent - calculated on the formulation.

9. The method of Claim 6, wherein said cosmetic formulations further contain at least one primary and/or secondary sun protection filter.

10. Carnosine compounds according to formula (I) as medicaments for preventing, mitigating and/or treating disorders of human skin or related dysfunctions induced by enhanced expression of PTGS2.

11. Compounds for use according to Claim 10, wherein said compound is L-Carnosine.

12. Compounds for use according to Claim 1, wherein said skin disorders are selected from inflammation, ageing and/or skin cancer.

13. Compounds for use according to Claim 1, wherein said related dysfunctions are selected from fever and/or pain. 14. Compounds for use according to Claim 1, wherein said carnosines are formulated in a cream, a lotion, an ointment or a spray.

15. A carnosine compound according to formula (I) in which R1 stands for H or CH3 and R2 for H or COOH, for use as a medicament for preventing, mitigating or treating PTGS2-induced disorders of human skin and related dysfunctions

A method for preventing, mitigating and/or treating PTGS2-induced skin disorders and related dysfunctions

AREA OF INVENTION

[0001] The present invention refers to the area of cosmetics and concerns a non-therapeutic method for treating specific skin disorders and related dysfunctions and a medicament for treating such disorders and dysfunctions.

BACKGROUND OF THE INVENTION

[0002] Prostaglandin-endoperoxide synthase 2 (PTGS2), also known as cyclooxygenase 2 (COX-2), is an inflammatory enzyme involved in prostaglandin biosynthesis. It is strongly induced by many stimuli including growth factors, cytokines and solar radiation, resulting in the production of prostaglandins during inflammatory processes. Therefore, inhibition of PTGS2 represent one way for preventing disorders of human skin, such as inflammation, aging and carcinogenesis, but also pain and fever, particularly - but not limited to - dysfunctions induced by solar radiation.

[0003] Various PTGS2 inhibitors are known from cancer and osteoarthritis research, such as:

Celecoxib (Celebrex) Rofecoxlb (Vloxx) Valdecoxlb (Bextra)

Etoricoxib (Arcoxla) Lumlracoxlb (Prexlge) [0004] All of these ingredients are pharmaceuticals that are subject to strict dosage regulations and therefore require a prescription. These substances are therefore not suitable for use in cosmetics.

RELEVANT PRIOR ART

[0005] From a study by Aruoma et al. in 1989 it is known that carnosine is able to scavenge ROS ("reactive oxygen species") [Biochem. J. 264(3), p. 863-869 (1989)]. Carnosine, according to a report by Babizhayev et al, can further scavenge alpha-beta-unsaturated aldehydes released during peroxidation of cell membrane fatty acids under oxidative stress [Membrane & Cell Biol. 12(1), pp. 89-99 (1998)].

[0006] EP 1310238 A2 (BASF) discloses cosmetic or dermatological photoprotective compositions comprising at least one cholesteric-liquid crystalline component reflecting in the infrared wavelength range from 750 nm to 2500 nm, at least one filter substance protecting against radiation in the ultraviolet radiation range from 280 nm to 449 nm, and at least one cosmetically acceptable carrier, and the use thereof. In addition, antioxidants, such as carnosine, may further be present.

[0007] EP 1388339 A1 (VISA) discloses the use of carnosine to control the aging process by reversing, preventing and reducing wrinkling of the skin. Carnosine is described as an antiwrinkle agent that also protects against glycosylation and chronic inflammation, among other effects.

[0008] EP 1591105 A1 (STADA) relates to cosmetic and pharmaceutical preparations containing an antioxidant that protects the skin from IR radiation. Among the numerous possible antioxidants, carnosine is mentioned. Further, the compositions to be topically administered comprise one or more inorganic and/or organic UV filters.

[0009] EP 2545898 A1 (COTY) discloses compositions that protect the skin from damage by IR rays, comprising plant extracts, vitamins, ruby powder, mica and titanium dioxide. Section [0026] also mentions carnosine as another suitable excipient, but as a free radical scavenger.

[0010] WO 1990 006102 A1 (PEPTIDE TECHNOLOGY) also concerns the anti-oxidative power of carnosine compounds. Mainly it is disclosed that the found compounds reduce or prevent collagen crosslinking in the skin and/or damage to DNA skin cells, especially caused by aging or UV radiation. The application is topical or by injection.

[0011] WO 2010 124817 A2 (KPSS) relates to conditioning agents based, among others, on dipeptides such as carnosine for hair.

[0012] WO 2014 128228 A2 (SYMRISE) demonstrates the suitability of carnosine for protection against IR radiation on the basis of inhibition of MMP expression. EP 3389621 A1 (SYMRISE) refers to emulsions comprising carnosine and UV filters. [0013] US 2006 216251 A1 (TRACIE MARTIN) relates to topical compositions containing carnosine, carnitine and a lipoic acid for the treatment and protection of the skin, in particular with respect to signs of skin aging related to chrono-ageing or photo-ageing, which is directed to wrinkles, skin weakening, and loss of elasticity. The treatment directed to hyperpigmentation is also disclosed. The influence of carnosine on MMP-1 inhibition is also pointed out.

[0014] KR 2004 0074795 A (A E KYUNG) discloses that 0.1 to 2 wt.-percent of carnosine in cosmetic compositions exhibit an antioxidant effect on the skin and increase collagen synthesis.

OBJECT OF THE INVENTION

[0015] Therefore, it has been the object of the present invention identifying a cosmetic active ingredient that is capable of inhibiting the expression of PTGS2 in order to prevent, mitigate or treat skin from disorders and/or dysfunctions induced by increased expression of PTGS2 such as inflammation, ageing, skin cancer, but also fever and pain.

BRIEF DESCRIPTION OF THE INVENTION

[0016] A first object of the present invention refers to a non-therapeutic method for preventing, mitigating or treating PTGS2-induced disorders of human skin and related dysfunctions, comprising or consisting of the following step: applying a working amount of a carnosine compound according to formula (I) in which R1 stands for H or CH3 and R2 for H or COOH, to human skin

[0017] Surprisingly it was found that treatments with well-known cosmetic ingredients of the carnosine-type, particularly using L-Carnosine, in amounts ranging from about 0.01% to about 5 wt.-percent, and preferably from about 0.1 to about 2.5 wt.-percent counteract the effect particularly of solar simulated radiation (SSR) gene expression of PTGS2, reducing the expression level of this gene. Also at protein level, the examples confirm a PTGS2 induction following SSR exposure and the protective effect of L-Carnosine treatment reducing the PTGS2 protein level in human skin. Therefore, carnosines in general and L-Carnosine in particular represent effective agents for prevention, mitigation and/or treatment of disorders of human skin, such as inflammation, ageing and skin cancer and related dysfunctions like for example fever and pain.

Carnosine compounds

[0018] Carnosines are basically known compounds which are accessible by the usual methods of organic chemistry. Preferably, these are group of substances formed by carnosine, L- carnosine, D-carnosine, D/L-carnosine, anserine, D-anserine, L-anserine as well as L- anserine*HNO3 and mixtures thereof. According to the invention, salts of the compounds of formula (I) are preferably understood as salts of the compounds of formula (I) with mineral acids, and particularly preferably salts of formula (II):

* n A, where n is 1, 2 or 3 and A is HCI or HNO3 and R1 is H or CH3 and R2 is H or COOH. The preferred carnosine is L-Carnosine, a dipeptide consisting of B-alanine and histidine.

COSMETIC FORMULATIONS

[0019] In a preferred embodiment the carnosine compounds applied to human skin as a component of a cosmetic skin or sun care formulation, which can be for example a cream, a lotion or a spray. Said carnosine compounds can be present in said cosmetic formulations in amounts of from about 0.01 to about 5 wt.-percent, preferably from about 0.1 to about 2.5 wt. -percent - calculated on the formulation.

[0020] These formulations may contain antidandruff agents, irritation-preventing agents, irritation-inhibiting agents, antioxidants, adstringents, perspiration-inhibiting agents, antiseptic agents, ant-statics, binders, buffers, carrier materials, chelating agents, cell stimulants, cleansing agents, care agents, deodorizing agents, antiperspirants, softeners, emulsifiers, enzymes, essential oils, fibres, film-forming agents, fixatives, foam-forming agents, foam stabilizers, substances for preventing foaming, foam boosters, gelling agents, gel-forming agents, hair care agents, hair-setting agents, hair-straightening agents, moisture-donating agents, moisturizing substances, moisture-retaining substances, bleaching agents, strengthening agents, stain-removing agents, optically brightening agents, impregnating agents, dirtrepellent agents, friction-reducing agents, lubricants, moisturizing creams, ointments, opacifying agents, plasticizing agents, covering agents, polish, gloss agents, polymers, powders, proteins, re-oiling agents, abrading agents, silicones, hair promotion agents, cooling agents, skin-cooling agents, warming agents, skin-warming agents, stabilizers, UV-absorbing agents, UV filters, detergents, thickeners, vitamins, oils, waxes, fats, phospholipids, saturated fatty acids, mono- or polyunsaturated fatty acids, oc-hydroxy acids, poly hydroxyfatty acids, liquefiers, dyestuffs, colour-protecting agents, pigments, odoriferous substances, polyols, surfactants, electrolytes, organic solvents or silicone derivatives and the like as additional auxiliaries and additives.

[0021] Preferably, the formulations further contain at least one, more preferably two, three, four or more primary and/or secondary sun protection factors, such as for example UVA and UVB filters and antioxidants (different from carnosines).

Surfactants

[0022] Preferred auxiliaries and additives are anionic and/or amphoteric or zwitterionic surfactants. Non-ionic and cationic surfactants can be also present in the composition. Suitable examples are mentioned along with the paragraph dealing with emulsifiers.

[0023] Typical examples for anionic and zwitterionic surfactants encompass: Almondami- dopropylamine Oxide, Almondamidopropyl Betaine, Aminopropyl Laurylglutamine, Ammonium C12-15 Alkyl Sulfate, In addition, German patent DE 4308794 C1 describes a process for the production of solid esterquats in which the quaterni- sation of triethanolamine esters is carried out in the presence of suitable dispersants, preferably fatty alcohols.

Other esterquats are those of which the acyl component derives from dicarboxylic acids like malonic acid, succinic acid, maleic acid, fumaric acid, glutaric acid, sorbic acid, pimelic acid, azelaic acid, sebac- ic acid and/or dodecanedioic acid, but preferably adipic acid. Overall, esterquats of which the acyl component derives from mixtures of monocarboxylic acids containing 6 to 22 carbon atoms, and adipic acid are preferably used. The molar ratio of mono and dicarboxylic acids in the final esterquat may be in the range from 1 :99 to 99:1 and is preferably in the range from 50:50 to 90:10 and more particularly in the range from 70:30 to 80:20. Besides the quater- nised fatty acid triethanolamine ester salts, other suitable esterquats are quaternized ester salts of mono-/dicarboxylic acid mixtures with diethanolalkyamines or 1,2-dihydroxypropyl dialkylamines. The esterquats may be obtained both from fatty acids and from the corresponding triglycerides in admixture with the corresponding dicarboxylic acids. One such process, which is intended to be representative of the relevant prior art, is proposed in European patent EP 0750606 B1. To produce the quaternised esters, the mixtures of mono- and dicarboxylic acids and the triethanolamine - based on the available carboxyl functions - may be used in a molar ratio of 1.1 :1 to 3:1. With the performance properties of the esterquats in mind, a ratio of 1.2:1 to 2.2:1 and preferably 1.5:1 to 1.9:1 has proved to be particularly advantageous. The preferred esterquats are technical mixtures of mono-, di- and triesters with an average degree of esterification of 1.5 to 1.9.

Superfatting agents and consistency factors

Enzyme inhibitors

Odour absorbers and antiperspirant active agents

[0067] Suitable odour absorbers are substances which are able to absorb and largely retain odour-forming compounds. They lower the partial pressure of the individual components, thus also reducing their rate of diffusion. It is important that perfumes must remain unimpaired in this process. Odour absorbers are not effective against bacteria. They comprise, for example, as main constituent, a complex zinc salt of ricinoleic acid or specific, largely odourneutral fragrances which are known to the person skilled in the art as "fixatives", such as, for example, extracts of labdanum or styrax or certain abietic acid derivatives. The odour masking agents are fragrances or perfume oils, which, in addition to their function as odour masking agents, give the deodorants their respective fragrance note. Perfume oils which may be mentioned are, for example, mixtures of natural and synthetic fragrances. Natural fragrances are extracts from flowers, stems and leaves, fruits, fruit peels, roots, woods, herbs and grasses, needles and branches, and resins and balsams. Also suitable are animal products, such as, for example, civet and castoreum. Typical synthetic fragrance compounds are products of the ester, ether, aldehyde, ketone, alcohol, and hydrocarbon type. Fragrance compounds of the ester type are, for example, benzyl acetate, p-tert-butylcyclohexyl acetate, linalyl acetate, phenylethyl acetate, linalyl benzoate, benzyl formate, allyl cyclohexylpropionate, styrallyl propionate and benzyl salicylate. The ethers include, for example, benzyl ethyl ether, and the aldehydes include, for example, the linear alkanals having 8 to 18 carbon atoms, citral, citron- ellal, citronellyloxyacetaldehyde, cyclamen aldehyde, hydroxycitronellal, lilial and bourgeonal, the ketones include, for example, the ionones and methyl cedryl ketone, the alcohols include anethole, citronellol, eugenol, isoeugenol, geraniol, linaool, phenylethyl alcohol and terpineol, and the hydrocarbons include mainly the terpenes and balsams. Preference is, however, given to using mixtures of different fragrances which together produce a pleasing fragrance note. Essential oils of relatively low volatility, which are mostly used as aroma components, are also suitable as perfume oils, e.g. sage oil, camomile oil, oil of cloves, melissa oil, mint oil, cinnamon leaf oil, linden flower oil, juniperberry oil, vetiver oil, olibanum oil, galbanum oil, labda- num oil and lavandin oil. Preference is given to using bergamot oil, dihydromyrcenol, lilial, lyral, citronellol, phenylethyl alcohol, oc-hexylcinnamaldehyde, geraniol, benzylacetone, cyclamen aldehyde, linalool, boisambrene forte, ambroxan, indole, hedione, sandelice, lemon oil, mandarin oil, orange oil, allyl amyl glycolate, cyclovertal, lavandin oil, clary sage oil, p- damascone, geranium oil bourbon, cyclohexyl salicylate, Vertofix coeur, iso-E-super, Fixolide NP, evernyl, iraldein gamma, phenylacetic acid, geranyl acetate, benzyl acetate, rose oxide, romilat, irotyl and floramat alone or in mixtures.

EXAMPLES

EXPERIMENTAL DESIGN

[0092] In order to evaluate the efficacy of L-Carnosine on protecting skin from Solar Simulated Radiation (SSR), gene expression and protein level of PTGS2 were analysed. -Ex vivo human skin biopsies were topically treated with L-Carnosine at different concentrations (0.2% and 2.2%), and daily exposed to 250 J/cm 2 of SSR. Gene expression and protein modulation were assessed respectively at day 2 and day3. -The radiation dose was selected following the literature [11 -13], and represent the approximative amount of solar light acquirable under natural conditions by sunbathing.

Tissue culture

[0093] Human skin samples were obtained from plastic surgery of healthy patients (aged between 25 and 60), and not intensive sun exposed. The informed consent was obtained from patients and retained by clinic. For the studies, twelve 8 mm diameter punch biopsies were used for each treatment. The skin samples were cultured in modified Williams' E medium at air-liquid interface and, maintained in incubator at the following conditions: 37°C, 5% CO2 and 95% humidity.

Treatments

[0094] For these studies L-Carnosine was dissolved in vehicle (DMSO/PBS 1 :25) at the following concentration 0.2% and 2.2%. Treatments were applied topically and renewed daily as follows: skin biopsies were cleaned with sterile cotton buds and 4 pl of treatment were applied on the biopsy's epidermis. All treated and the control samples were covered with 6 mm diameter delivery membranes.

Solar Simulated Radiation (SSR)

[0095] Skin irradiation was performed daily, with Suntest XLS+ (Ametek Inc., USA) equipped with the provided IR filter and a DAY LIGHT filter, to obtain an emission spectrum between 300 and 800 nm. Every day the skin samples were irradiated with a SSR dose of 250 J/cm 2 . Control samples were sham-irradiated under the same conditions.

Gene expression analysis [0096] Human skin biopsies were collected in cryo-vial containing a commercial RNA stabilization solution (RNAIater, Merck KGaA, Germany) and stored at -80°C. Total RNA was extracted using a commercial kit (Rneasy mini-kit, Qiagen GmbH, Germany) according to the manufacturer's instructions. The expression level of selected genes was determined using Real-time RT-PCR method with custom designed gene array (TaqMan® Array 96-Well Fast Plates, Thermo Fisher Scientific Inc., USA). Gene expression changes were determined in experimental samples compared to the not irradiated control, and values were expressed in terms of fold-change versus control (fold-change control = 1; fold-change up-regulated level >2).

IHC staining

[0097] Skin samples were collected (day 3), fixed in formalin and paraffin embedded. Skin sections were immuno-stained with PTGS2 antibody (Thermo Fisher Scientific Inc., USA). The images of the skin sections were acquired, and the analysis was performed through Image-J application (NIH, USA).

Example 1

PTGS2 gene expression level

Gene expression changes were determined in three donors. For each donor, experimental sample were compared to the not irradiated vehicle (control) and values were expressed in terms of fold-change versus control (fold-change control = 1.0; fold-change up-regulated level >2, statistical significance A > 0.5). The results are shown in Tables 1 and 2.

Summary

The results indicate that L-carnosine reduces the expression of PTGS2 and provides a photo- protective effect to cells. The results are further illustrated by Figures 1a-1c. Example 2

The results, indicate that L-Carnosine is capable to efficiently reduce the SSR-induced induction of the PTGS2 gene and the PTGS2-protein. This result indicates that L-carnosein reduces PTGS2 induced skin disorders and related dysfunctions.
