Abstract:
The present invention addresses the problem of providing a method whereby it becomes possible to separate hardly separable impurities having similar physical properties to those of a desired antibody fragment and it also becomes possible to produce the desired antibody fragment with high efficiency and high purity. The method for producing an antibody fragment according to the present invention is characterized in that the antibody fragment contains a CH1 domain but does not contain an Fc domain, and is also characterized by comprising the steps of: preparing a liquid sample that contains the antibody fragment but does not contain an Fc fragment; bringing the liquid sample into contact with an affinity separation matrix comprising an insoluble carrier and protein G, a domain thereof or a mutant of protein G or the domain immobilized, as a ligand, on the insoluble carrier to cause the adsorption of the antibody fragment onto the affinity separation matrix; washing the affinity separation matrix to remove impurities; and separating the antibody fragment from the affinity separation matrix.
