Abstract:
The present invention provides a method whereby at least two DNA materials are assembled and DNA fragments having a desired base sequence are produced. This method comprises: a step (1) in which at least two DNA materials are linked and a precursor DNA fragment is obtained that has part of a desired base sequence and a linking base sequence alternating therein; and a step (2) in which the precursor DNA fragment is treated using a restriction enzyme, the linking base sequences are removed, and a DNA fragment having the desired base sequence is obtained. The base sequences recognized by the restriction enzyme are arranged in the precursor DNA fragment such that, when the base sequences are treated using the restriction enzyme and the precursor DNA fragment is cleaved, the linking base sequences are removed and the parts of the desired base sequences that are adjacent via the linking base sequences can be linked at the cleaved sites.
