Abstract:
The present disclosure provides a method for producing a nucleic acid molecule. In detail, the present disclosure realizes process shortening and dramatic efficiency in ligation by using nucleic acid splints. Specifically, the present disclosure is a method for producing a polynucleotide molecule having a specific sequence, the method including 1) a step of producing two or more polynucleotide fragment molecules including a part of said specific sequence and 2) a step for connecting said two or more polynucleotide fragment molecules in the presence of two or more nucleic acid splints. Said specific sequence is formed when all the sequences of said two or more polynucleotide fragment molecules are assembled. Said nucleic acid splits are substantially complementary to said polynucleotide molecules. When at least two of said nucleic acid splints align with said specific sequence, a sequence in which there is substantially no deletion is obtained.
