Abstract:
Disclosed in the present invention are a method for purifying a reversibly blocked deoxyribonucleotide triphosphate and a sequencing method, the method for purifying a reversibly blocked deoxyribonucleotide triphosphate comprising: using an adenylate cyclase to treat a 3' reversibly blocked dATP, and/or using guanylate cyclase to treat a 3' reversibly blocked dGTP. When used in sequencing, a reversibly blocked dNTP which has been treated may lead to lower phase shifting, especially for a pre-determined phase, so as to improve sequencing quality.
