Abstract:
Disclosed in the present invention are a method and a reagent for enrichment of circulating tumor DNA (ctDNA). The method comprises the following steps: mixing a water phase and an oil phase, oscillating to prepare a reaction system of emulsion PCR and performing the emulsion PCR amplification, wherein the water phase comprises the peripheral blood plasma DNA as the template DNA, a forward primer and a reverse primer, dNTPs, PCR buffer and DNA polymerase, adapter sequences are connected to the two ends of the peripheral blood plasma DNA, and the forward primer and the reverse primer are complementarily paired to the adapter sequences of the two ends respectively; separating the water phase and the oil phase after the emulsion PCR amplification to obtain the PCR amplification product of the water phase; capturing the circulating tumor DNA in the PCR amplification product of the water phase by using the probes specifically binding to the circulating tumor DNA. The method of the present invention is capable of capturing the peripheral blood plasma ctDNA effectively and performing a single molecule high-fidelity ultramicro parallel amplification to provide adequate ctDNA used for the subsequent sequencing detection.
