Abstract:
Provided are a method and a reagent for the separation, enrichment and nucleic acid extraction of pathogenic microorganisms in blood. The method comprises: firstly, adding a blood selective lysis solution A and a pathogenic microorganism settling aid to a blood sample to selectively lyse blood cells, and settling pathogenic microorganisms by means of high-speed centrifugation; then adding a blood selective lysis solution B to selectively lyse the blood sample again, and then adding a washing solution, and uniformly mixing and then washing same; and finally adding a nucleic acid extraction reagent, centrifuging and enriching the pathogenic microorganisms, and then performing lysis in combination with a physical method to obtain a pathogenic microorganism nucleic acid sample. The nucleic acid extraction method can quickly and accurately extract the nucleic acid of an infectious target pathogen, which can meet a molecular biology detection requirement, from a blood sample not greater than 10 mL, and the nucleic acid extraction step is simple, complex processing equipment is not needed, and the obtained nucleic acid can be used for various downstream detections in molecular biology.
