Abstract:
The present invention relates to a method for sensing and quantifying ε-caprolactam by using a redesigned gene circuit and, more particularly, to a method for analyzing and quantifying ε-caprolactam by using a gene circuit which can sense ε-caprolactam which is used as a precursor of nylon-6. The redesigned gene circuit recognizing ε-caprolactam according to the present invention allows a regulatory gene of ε-caprolactam to induce the expression of a fluorescent protein in proportion to an amount of ε-caprolactam. Thus, it is possible to rapidly sense and quantify ε-caprolactam. Therefore, the redesigned gene circuit can be used for the purpose of quantitatively analyzing ε-caprolactam and analyzing the activity of an enzyme so as to biosynthesize ε-caprolactam. Through the present invention, a trace amount - i.e., about 0.1 μm of ε-caprolactam - can be sensed by means of high-sensitivity, and analysis in cell units, which was not possible in the related art, is made possible. Thus, the present invention can be effectively used for a screening process of a novel gene and strain which produce ε-caprolactam from a microorganism metagenome library by using high-throughput screening.
