Abstract:
Disclosed in the present invention is a method for synthesizing L-aspartic acid by means of the whole cell catalysis of maleic acid by coupling two enzymes, which relates to the field of bio-engineering. In the present invention, a recombinant strain that coupledly expresses a cis-trans isomerase of maleic acid and L-aspartate lyase is constructed, and the recombinant strain is further modified and optimized to obtain a recombinant bacterial strain which may perform whole cell high-conversion rate catalysis of maleic acid to generate L-aspartic acid. The recombinant strain may produce L-aspartic acid by using maleic acid which is relatively inexpensive; after reacting for 40-120 min, the maleic acid has completely reacted, and almost none of the intermediate product of fumaric acid has accumulated, wherein the conversion rate may reach 98% or higher.
