Abstract:
The present invention relates to a use of a reference cell for testing the expression level of a pluripotency gene, wherein the reference cell is selected from cells having a low and stable expression level of the pluripotency gene. The reference cell is selected from HFF cells. The pluripotency gene is selected from OCT4 and NANOG. The present invention further relates to a primer and a probe for OCT4 and NANOG, and a method for testing the expression level of a pluripotency gene. The present invention further relates to a use of the testing method in hPSC residue level testing and hPSC differentiation process characterization. The method according to the present invention is easy to standardize, and can provide a unified standard for the testing of the expression level of a cell pluripotency gene.
