Document Type and Number:
WIPO Patent Application WO/2010/115249
Kind Code:
A1
Abstract:
Subclinical mastitis treatment method in lactating animals using photo dynamic therapy is an innovative therapeutic method to mastitis treatment in lactating animals that consists in an optical treatment within the animal's mammary gland, maximizing with more efficiency the bacteria death with the administration of photosensitive agent. Thus, it was used low-energy laser (LEDs) emission equipments with no thermal effect, already existents, substituting the usual tips by acrylic ones with specific dimensions that enable its penetration on the animal teat orifice to guide the light emission to reach the gland internally irradiating in three distinct deep levels with the energy density properly adjusted to each of them in only one application direct on the infection focus together the introduction of a photosensitive agent into the teat by a urethral probe, that stimulated by low-energy laser (LEDs) irradiation, has absorbed the radiation an has transmitted these captured energy to the cells, triggering the desired photosensitization effect, ie the death of microorganisms by light in presence of a photosensitive agent.

Description:
METHOD FOR TREATMENT OF SUBCLINICAL MASTITIS IN LACTATING ANIMALS USING PHOTODYNAMIC THERAPY

Field of Invention

The present application of Invention Privilege belongs to the therapeutic procedures field having the objective of defining an innovative subclinical mastitis treatment method in lactating animals using "Photo Dynamic Therapy" (PDT), that consists on applying a photosensitive agent together laser irradiation on the infected mammary quarter, using an already existing low- energy laser emission equipment (LEDs), that can be adapted to any acrylic instrument tip, allowing its introduction from the animal teat orifice till the desired location, so the radiation reaches the entire glandular part of the infected teat in three different levels in only one application with an established nism to death without loss or excessive exposure.

The use of this innovative subclinical mastitis treatment method in lactating animals through Photo Dynamic Therapy (PDT) makes short, with total elimination of the microorganism, these infections duration, a fundamental importance aspect in control and prevention programs of mastitis during lactation; eliminates the effective milk secretion reduction, as the milked milk disposal, both originated by the infection by microorganism presence and by the antibiotics residues applied on usual treatments. History of Invention

Studies show that bovine mastitis is currently one of the most serious problems found on dairy husbandry, with high prevalence in flocks all around the world that is being studied since the eighteenth century and even today with a lot of studies about this pathology, it still is the disease with highest economical impact over milk production.

Subclinical mastitis causes a silent reduction of the milk capacity secretion of the affected mammary quarter and it can cause a huge impact, since the use of veterinary antimicrobial (antibiotics) chemical substances increases.

The conventional treatment is done with intra mammary antibiotics application and, in last cases, when all the animal organism is affected, the application is parenteral (intramuscular).

The concern with a growing antibiotics residues presence on milk generates a search for alternative methods to the classical approach of this substances, for example the use of homeopathic and biotherapy medicines prepared from bacteria, fungi, viruses, pathological secretions or excretions, mites, worms, etc. either animal or vegetal.

State of Technical Disabled Points

1 - The conventional long duration treatment to microorganism eradication with antibiotics application, either via mammary or injectable, compromises the milk quality by the antimicrobial chemical substance (antibiotics) presence, coming from the treatment, making necessary to discard the milk for up to 10 days.

2 - The subclinical mastitis treatment with antibiotics, in addition to present its residues on milk, can also produce residues on other animal origin products intended to human consumption and thus indirectly causing bacteria resistance on humans.

3 - The homeopathic medicines use, although the results are positive, the treatment has a long duration compromising the milk production.

4 - The biotherapy medicines application was effective on mastitis preventive treatment, but with a long duration on the infection treatment, also compromising the milk production.

Invention summary

Thinking about these problems that, after much research and studies, the inventors have created and developed an innovative subclinical mastitis treatment method, that eliminates the effective milk production reduction and disposal, meeting the increasingly stringent requirement of national and international institutions about the antimicrobial residues absence, through Photo Dynamic Therapy already known and used in humans on skin, intestine and other cancer treatments with low energy density and by dentists to eliminate any bacteria that may stay after a treatment before covering the tooth with high energy density, however never used on mastitis treatment.

The Photo Dynamic Therapy is the use of a low-energy laser producing a photochemical effect when applied against living tissue and in bacteria's case is maximized with the pigment use (photosensitive agent) plus the laser irradiation. The laser therapy was used in women on acute mastitis treatment applying a high power laser. Because animal's skin is very thick, a factor that decreases laser radiation absorption, the great innovation of Photo Dynamic Therapy to treat mastitis in lactating animals is the topic within the animal's mammary gland, as a more efficient method that maximizes the bacteria death in association the photosensitive agent administration.

Thus, it was used low-energy laser (LEDs) emission equipments with no thermal effect, already existents, substituting the usual tips by acrylic ones with 100 mm in length and 5 mm in diameter, that enables the penetration on the animal teat orifice to guide the light emission to reach the gland internally. Laser was irradiated in three distinct deep levels with the energy density properly adjusted in 200 J/ cm 2 in each one, never tested in animals before, it enabled only one direct application on infection focus. It was also used an introduction of 1 ml of photosensitive agent, Toluidine blue, in teat by urethral probe with a 2,5% concentration, that stimulated by laser irradiation, transmits the captured energy to the cells, triggering the desired photo photosensitization effect, ie the death of microorganisms by light in the presence of a photosensitive agent.
