Abstract:
The present invention discloses a method for visually marking a genomic site and an application thereof in the detection of cell and organism aging levels. The present invention integrates thioredoxin (TRX) at the C-terminus of a transcription activator-like effector (TALE) protein to create a visual genome marking tool, TTALE, for marking telomeres, centromeres, rDNA, and other genomic repetitive sequences and encoding loci in different types of human cells. The present invention also discloses a kit and a method for detecting the number of rDNA copies in the genome of human cells to detect or assist in detecting the aging level of a cell or an organism to be tested.
