Abstract:
A microneedle patch for enhancing an accumulation of protoporphyrin IX in a solid tumor and a preparation method therefor. The method comprises the steps of: dissolving albumin in a Dulbecco's culture medium, using the albumin as a template, and adding CuCl2 and CaCl2 for mineralization to obtain a CCP nanoparticle; by means of a one-step carbodiimide coupling method and an electrostatic adsorption method, loading CAT and 5-ALA to the surface of the CCP nanoparticle, respectively, to obtain a CCPCA nanoparticle; and loading the CCPCA nanoparticle and a microneedle matrix solution in a microneedle mold, drying under vacuum, and demolding to obtain a microneedle patch for enhancing an accumulation of PpIX in a solid tumor. According to the microneedle patch, by means of an integrated nano calcium phosphate carrier and a transdermal administration mode, an accurate delivery of CAT and 5-ALA is achieved, and PpIX and O2 levels in a tumor cell are increased. Meanwhile, the guidance of real-time bimodal PpIX fluorescence imaging and photoacoustic blood oxygen imaging is provided, and the photodynamic therapeutic effect on different cancers is improved.
