Abstract:
A miniaturized adaptive optical two-photon fluorescence imaging system and method. The imaging system comprises: a laser source device (1), for outputting excitation light; a miniature probe device (2), for receiving the excitation light outputted from the laser source device (1), exciting a tissue surface in a live sample by the excitation light to generate a fluorescent signal and, while the live sample is in a free-movement state, performing, according to an average wavefront distortion distribution of isoplanatic regions of an imaging field of view of the tissue surface in the live sample, wavefront correction with respect to the isoplanatic regions; a wavefront detection device (3), having a signal input port operably connected to a signal output port of the miniature probe device (2) and being for, while the live sample is fixed, receiving the fluorescent signal outputted from the miniature probe device (2) and detecting the average wavefront distortion distribution; a de-scanning member (4), arranged on an optical path between the miniature probe device (2) and the wavefront detection device (3), for compensating in real time an isoplanatic region off-axis effect generated by the miniature probe device (2) before the wavefront detection device (3) detects the average wavefront distortion distribution; and a fluorescent imaging device (5), having an input signal port operably connected to a signal output end of the miniature probe device (2), for collecting the wavefront-corrected fluorescent signal outputted from the miniature probe device (2) to complete imaging of the tissue surface in the live sample. The imaging system and the imaging device achieve a high temporal and spatial resolution as well as large field-of-view imaging of deep biological tissues in freely moving animals.
