Abstract:
The present invention relates to an mRNA or mRNA construct including: (a) a region encoding a protein or peptide of interest; (b) a β-globin UTR region at the 5'end; and (c) a region at the 3'end in which BGH, BGH-IRES, or IRES-BGH is inserted. An mRNA or mRNA construct according to the present invention improves intracellular expression or expression stability, and intracellular persistence of a chimeric antigen receptor or chimeric antigen receptor protein (CAR protein). In addition, the mRNA or mRNA construct increases the in vivo safety of the chimeric antigen receptor, and increases the ability thereof to kill cancer cells, and thus can be effectively used in anticancer pharmaceutical compositions and anticancer immunotherapy targeting solid tumors.
