Abstract:
A multi-layer totally enclosed microfluidic chip, comprising: a base layer (1), a reaction layer (2), an adhesive layer (3), and a sample-adding layer (4), which are sequentially attached and fixed from bottom to top, wherein a press cap (5) is clamped at the top of the sample-adding layer (4) in a snap-fit manner; the middle of the reaction layer (2) is provided with a vertically penetrating sample injection channel (21), vertically penetrating reaction cells (22) being provided around the sample injection channel (21), the bottom surface is provided with inflow channels (23) respectively communicating the sample injection channel (21) with the reaction cells (22), the top surface is provided with gas filter cells (24) around the reaction cells (22) and with the same number as the reaction cells (22), and the top surface is provided with outflow channels (25) respectively communicating the gas filter cells (24) with the reaction cells (22); the adhesive layer (3) is provided with conformal slots (31) corresponding to the sample injection channel (21), the reaction cells (22), the outflow channels (25) and the gas filter cells (24); the middle of the sample-adding layer (4) is provided with a vertically penetrating sample-adding cell (41) corresponding to the sample injection channel (21), and the sample-adding layer (4) is provided with vertically penetrating exhaust holes (42) corresponding to the gas filter cells (24); and a hydrophobic gas-permeable membrane (6), which closes the exhaust holes (42), is pressed between the press cap (5) and the sample-adding layer (4). A quantitative sample-adding device for use cooperatively with the multi-layer totally enclosed microfluidic chip comprises a quantitative sample cylinder (10), a quantitative pressing structure (9) and a liquid injection head (11).
