Abstract:
The present invention is used in the technical fields of molecular biology and clinical diagnostics. Disclosed is a method using a multi-layered centrifugal filter for exosome extraction. The method uses the multi-layered centrifugal filter device for exosome extraction and an exosome extraction kit consisting of an incubation buffer and protease K. The method comprises: using the incubation buffer in the kit and a suitable amount of the protease K to incubate, under room temperature, a sample for testing; then using a multi-layered centrifugal filter device on a matching centrifuge to perform centrifugation, mixing to homogeneity, then collecting a retained fluid in an ultrafiltration tube (2) to acquire the exosome. The method does not require large experimental apparatus other than a centrifuge. The method has low costs and is easy to operate, and requires a short operating time and can be used to perform parallel processing on a large volume of samples. A high purity exosome obtained through the process can satisfy a requirement for promoting large scale application in a clinical setting.
