Abstract:
A CRISPR nucleic acid testing method, comprising the following steps: step 1, treating samples; step 2, preparing a reaction system which comprises: crRNA complementary to a target sequence to be tested, tracrRNA, Cas14 enzyme and an enzyme buffer; step 3, adding a nucleic acid to be tested and a probe to a nucleoglucoprotein complex solution; and step 4, performing enzyme cleavage reaction, signal detection and result interpretation. This method can be used to the test of multiple types of nucleic acids and multiple sites such as single-stranded DNA, RNA, double-stranded DNA and single-site mutant CRISPR nucleic acids, and has high design flexibility, good specificity, convenience, rapidness and low cost.
