Abstract:
The present invention provides a method for manufacturing nucleic acid-encapsulating empty particles of the adeno-associated virus (AAV), the method comprising the following steps: (1) a step in which linear nucleic acid fragments are prepared, the nucleic acid fragments comprising (i) the sequence of the A region of an AAV inverted terminal repeat sequence (ITR), (ii) the sequence of the A' region of the AAV ITR, and (iii) the sequence of the D region and/or the sequence of the D' region of the AAV ITR; (2) a step in which the nucleic acid fragments from (1) are introduced into cells that generate AAV empty particles; and (3) a step in which the cells from (2) are cultured.
