Abstract:
Provided are an optical detection method and an optical detection apparatus that quickly and accurately detect a very small target substance such as an antigen with high sensitivity by using an enhanced electric field. This optical detection apparatus is provided with: one or more light irradiation units; a detection plate that has a laminate structure formed by stacking a light transmissive substrate, a meal layer or a semiconductor layer, and a light transmissive dielectric layer in this order from the back surface toward the front surface; a prism that has a plurality of light incident surfaces and that is optically bonded to the back surface side of the detection plate, at least two of the light incident surfaces being different in incident surface angles formed between the light incident surfaces and the front surface of the detection plate; a sample holding unit; and a photodetection unit that detects an optical signal from the detection area of a sample and that is arranged on the front surface side of the detection plate. The optical detection apparatus is arranged in such a manner that light from the light irradiation unit enters the light incident surfaces of the prism at one fixed angle to the front surface of the detection plate, and the light passing through the prism is emitted from the back side surface of the detection plate under the condition of satisfying a full-reflection condition in the detection plate, and the optical signal is detected by the photodetection unit.
