Abstract:
This invention provides a microscopy technology that, using a scanning molecule-counting technique, makes it possible to detect light-emitting bodies or light-emitting particles that dynamically change in position in a thick sample. This optical microscopy technology detects light-emitting particles (LP) moving in a liquid by detecting light from said light-emitting particles (LP) via confocal microscope optics or multiphoton microscope optics. In a processing operation, light from a light-detection region (CV) is detected as said light-detection region (CV) is moved within each of a plurality of observation subregions (OsR) obtained by partitioning the observation region (ObR) of the microscope. This yields an individual signal corresponding to the light from each light-emitting particle (LP), and the position within the observation region (ObR) of the light-emitting particle (LP) corresponding to each detection signal is determined. The movement of the light-detection region (CV) within each observation subregion (OsR) may be performed a plurality of times in succession in the same direction and/or in two or more directions in each observation subregion (OsR).
