Provided are a primordial follicle activator and an application thereof in the preparation of a human ovarian cortex culture; the primordial follicle activator is composed of a mammalian target of Rapamycin (mTOR) signal pathway activator and a phosphoinositide 3-kinase (PI3K) signal pathway activator; the mTOR signal pathway activator comprises mTOR signal pathway activator I, and the mTOR signal pathway activator I is 50-200 μM of phosphate; the PI3K signal pathway activator comprises PI3K signal pathway activator II, and the PI3K signal pathway activator II is 100-1000 μM of 740 Y-P.
