Abstract:
The present invention deal with a process to obtain a tooth-paste and/or a mouth-wash containing antioxidants compounds obtained from plants that are rich in polyphenols, for the treatment or prevention of periodontal diseases where an increase in production of pro-oxidant compounds and/or for instant formation of large quantities of reactive oxygen species.

CLAIMS

"PROCESS TO OBTAIN TOOTH-PASTE AND/OR MOUTH-WASH (ORAL ANTISEPTICS), CONTAINING ANTIOXIDANT NATURAL OR SYNTHETIC, AND ANTIOXIDANT OBTAINED FROM PLANTS RICH IN POLYPHENOLS, USED IN THE PREVENTION AND/OR IN THE TREATMENT OF PERIODONTAL DISEASES IN WICH THERE ARE AN INCREASE OF PRO- OXIDANTS FACTORS AND/OR FORMATION OF LARGE QUANTITIES OF REACTIVE OXYGEN SPECIES."

1) Process for obtainment of tooth-paste containing dry residue and/or lyophilized of the hydro-alcoholic extract obtained from deccotion of fruits and/or stone of fruits of Euterpe oleracea, characterized by the following step. 1.1 During the manufacture of tooth-paste the dry residue or the lyophilized of hydro-alcoholic extract of fruits and/or stone of Euterpe oleracea are added in order to obtain tooth-paste containing from 0.1 to 100 mg of dry residue or lyophilized of the hydro-alcoholic extract of fruits and/or stone of fruits from Euterpe oleracea per gram of tooth-paste that can be used in the prevention and/or in the treatment of periodontal diseases.

2) Process for obtainment of mouth-wash containing dry residue and/or lyophilized of the hydro-alcoholic extract obtained from deccotion of fruits and/or stone of fruits of Euterpe oleracea, characterized by the following step. 2.1 During the manufacture of mouth-wash the dry residue or the lyophilized of hydro-alcoholic extract of fruits and/or stone of Euterpe oleracea are added in order to obtain a mouth-wash containing from 0.1 to 100 mg of dry residue or lyophilized of the hydro-alcoholic extract of fruits and/or stone of fruits from Euterpe oleracea per 100 ml of mouth-wash that can be used in the prevention and/or in the treatment of periodontal diseases. 3) Process for obtainment of tooth-paste containing the dry residue and/or the lyophilized obtained from red wine obtained from Vitis vinifera and/or Vitis labrusca, characterized by the following step.

3.1 During the manufacture of tooth-paste the dry residue or the lyophilized of red wine obtained from Vitis vinifera and/or Vitis labrusca grapes are added in order to obtain tooth-paste containing from 0.1 to 100 mg of dry residue or lyophilized of red wine per gram of tooth-paste that can be used in the prevention and/or in the treatment of periodontal diseases.

4) Process for obtainment of mouth-wash containing lyophilized and/or the dry residue of wine obtained from Vitis vinifera and/or Vitis labrusca grapes, characterized by the following step.

4.1 During the manufacture of mouth-wash the dry residue or the lyophilized of red wine obtained from Vitis vinifera and/or Vitis labrusca grapes are added in order to obtain mouth-wash containing from 0.1 to 100 mg of dry residue or lyophilized of red wine obtained from Vitis vinifera and/or Vitis labrusca grapes per 100 ml of mouth-wash that can be used in the prevention and/or in the treatment of periodontal diseases.

5) Process for obtainment of tooth-paste containing ascorbic acid, characterized by the following step. 5.1 During the manufacture of tooth-paste ascorbic acid is added in order to obtain tooth-paste containing from 0.1 to 100 mg of ascorbic acid per gram of tooth-paste that can be used in the prevention and/or in the treatment of periodontal diseases.

6) Process for obtainment of mouth-wash containing ascorbic acid, characterized by the following step.

6.1 During the manufacture of mouth-wash ascorbic acid is added in order to obtain mouth-wash containing from 0.1 to 100 mg of ascorbic acid per 100 ml of mouth-wash that can be used in the prevention and/or in the treatment of periodontal diseases. 7) Process for obtainment of tooth-paste containing tocoferol, characterized by the following step.

7.2 During the manufacture of tooth-paste tocoferol is added in order to obtain tooth-paste containing from 0.1 to 100 mg of tocoferol per gram of toothpaste that can be used in the prevention and/or in the treatment of periodontal diseases. 8) Process for obtainment of mouth-wash containing tocoferol, characterized by the following step.

8.1 During the manufacture of mouth-wash tocoferol is added in order to obtain mouth-wash containing from 0.1 to 100 mg of tocoferol per 100 ml of mouth-wash that can be used in the prevention and/or in the treatment of periodontal diseases.

"PROCESS TO OBTAIN TOOTH-PASTE AND/OR MOUTH-WASH (ORAL ANTISEPTICS), CONTAINING ANTIOXIDANT NATURAL OR SYNTHETIC, AND ANTIOXIDANT OBTAINED FROM PLANTS RICH IN POLYPHENOLS, USED IN THE PREVENTION AND/OR IN THE TREATMENT OF PERIODONTAL DISEASES IN WICH THERE ARE AN INCREASE OF PRO- OXIDANTS FACTORS AND/OR FORMATION OF LARGE QUANTITIES OF REACTIVE OXYGEN SPECIES."

I) Field of Invention The present invention deal with dental products and/or toothpaste and/or mouth-wash containing antioxidants obtained from plants rich in polyphenols, more precisely plants as Euterpe oleracea (acai), Vitis vinifera or Vitis labrusca, more particularly the use of those products, tooth-paste and/or mouth-wash, containing those anti-oxidants in the prevention and treatment of periodontal diseases where exist a large formation of reactive oxygen species.

II) Invention Antecedents

Data from the literature demonstrate that periodontal diseases, mainly those where inflammation is present, there is a large formation and release of products with pro-oxidant activity probably from microorganism present in the oral cavities (Akalin et al., Lipid peroxidation levels and total oxidant status in serum, saliva and gengival crevicular fluid in patients with chronic periodontitis. J Clin Periodontl 2007,34:558-65, - Wei et al., The investigation of glutathione peroxidase, lactoferrin, myeloperoxidase and interleukin-1 beta in gengival crevicular fluid: implication for oxidative stress in human periodontal diseases. J Periodontal Res. 2004, 39:287-93). Therefore utilization of toothpaste and/or toothpaste and/or mouthwash containing anti-oxidant compounds is a significant way to prevent and/or to treat periodontal diseases where an oxidative stress is present.

III) Summary of the Invention. The present invention refers to a process to obtain tooth-paste and/or mouthwash containing antioxidant products to be use to prevent and/or treat periodontal diseases where an oral oxidative stress is present. More precisely refer also of products as tooth-paste and/or mouth-wash solutions containing anti-oxidants compounds such as extracts obtained from plants rich in polyphenols as Euterpe oleracea (acai) and also from red wines obtained from Vitis vinifera or Vitis labrusca grapes, and also ascorbic acid or tocoferol.

IV) Condensed description that supports the production and utilization of toothpaste and/or mouth-wash for the prevention and treatment of periodontal diseases. The present invention is supported by scientific data obtained from the scientific literature that support the production and utilization of toothpaste and/or mouthwash on the prevention and/or treatment of periodontal diseases. Below we give a brief view of scientific data that supports the invention. The pathological oral process can be induced by the deleterious action of reactive oxygen species and/or pro-oxidant products that induce periodontal cellular lesions and therefore pathological alterations on the oral cavity. Therefore, utilization of tooth-paste and/or mouth-wash that contain compounds with anti-oxidant actions may avoid oral lesions and/or treat oral diseases induced by increase oral oxidative stress. More precisely, tooth-paste and/or mouth-wash will be manufactured having in its composition anti-oxidants as ascorbic acid, tocoferol, anti-oxidants obtained from plants rich in polyphenols, compounds that have anti-oxidant activities. More PARTICULARLY?????? Precisely, compounds with anti-oxidant activities may be obtained from fruits of plants as Euterpe oleracea (acai) or also from wines produced from Vitis vinifera and Vitis labrusca grapes. The data described above, were described not with the scope to limit, but wit the scope to illustrate the possibilities of realization of the present invention, with ambit is limited only by the claim annex.

V.A) Method to obtain the dried residue. Ivophilized or spray dried of the hydro alcoholic extract of the decoction of the whole fruits or decoction of the stone of the fruits of Euterpe oleracea. The stones of the fruit of Euterpe oleracea are obtained by elimination of the skin of the whole fruit. The fruits or the stones are washed and boiled in distilled water for 3 to 9 minutes to obtain the decoction of the whole fruits or from the stones. After boiling the decoction are minced and/or grinded, and then boiled for another 5 minutes e extracted with ethanol 95% in a proportion that varies from 1 :0.5 to 2:10, for instance 1:1. This mixture is again minced and/or grinded, and then macerated for a certain period of time (3 hours to 15 days) for instance 6 days at 4° C or at room temperature for instance 25° C under intermittent agitation. At the end of the maceration time the extract is filtered in a sieve with 0.1 to 1.0-mm pore, for instance 0.2 mm, being also filtered through gauze and finally filtered through a paper filter, for instance Whartman n.1. The semi-solid phase can be re-extracted on the same conditions already described for 1 to 3 times, for instance 2 times. The first liquid phase is kept inside a refrigerator at 4° C. The subsequent liquid phases are added to the first one. The liquid phase is concentrated in a low-pressure evaporator at a temperature of 35 to 65 ° C for instance 50° C and then lyophilized or spray dried.

IV.B) Method to obtain the dried residue, lyophilized or spray dried from wines obtained from Vitis vinifera and/or Vitis labrusca grapes. Red wines obtained from Vitis vinifera and/or Vitis labrusca grapes is initially concentrated in an evaporator at low pressure until the removal of all alcohol and then the dry residue is obtained by lyophilization and/or by spray dreyer. The dry residue, containing from 10 to 100 mg% of polyphenols is kept in a deep freezer until day of utilization in the manufacture of toothpaste and/or mouth-wash.

V Pharmacotechnical aspects of the preparation of toothpaste and/or mouthwash containing the Ivophilized obtained from the hydro alcoholic extract obtained from decoctions of fruit and stone of Euterpe oleracea, red wine and/or ascorbic acid or tocoferol. In order to obtain tooth-paste and/or mouth-wash containing antioxidants, during the process of manufacture of tooth-paste and mouth-wash the dry residue of hydro-alcoholic extract of fruits and stone from Euterpe oleracea, or dry residue of red wine obtained from Vitis vinifera or Vitis labrusca grapes or ascorbic acid or tocoferol will be added in various concentrations, with the objective to manufacture tooth-paste and/or mouth-wash with pharmacological properties in order to avoid development or to treat periodontal diseases where an oxidative stress is present, formation of large quantities of reactive oxygen species.
