Abstract:
Provided are a puromycin linker and an application thereof in synthesis of an in-vitro nucleic acid displayed peptide. The puromycin linker is a modified and unbranched single-stranded DNA; the single-stranded DNA comprises a first stretch of nucleotides and a second stretch of nucleotides; the 5' end of the first stretch of nucleotides is linked to the 5' end of the second stretch of nucleotides to form a linker structure containing two 3' ends; puromycin is modified at the 3' end of the first stretch of nucleotides; and the second stretch of nucleotides comprise, in an order of 5' to 3', an mRNA linking site and a reverse transcription site. A synthesis method for the puromycin linker is simple; the efficiency is up to 53%; the efficiency of obtaining key raw materials is greatly improved; and the system efficiency is further improved and synthesis costs of a cDNA-protein are reduced.
