Abstract:
The present application relates to a recombinant vector, and a construction method therefor and the use thereof. The method for constructing the recombinant vector comprises the following steps: constructing an sgRNA skeleton plasmid with a chemogenetic activation element; and inserting an sgRNA fragment into the sgRNA skeleton plasmid with the chemogenetic activation element to obtain a recombinant vector, wherein the sgRNA fragment is used for knocking out vGAT. According to the method for constructing the recombinant vector, the recombinant vector is obtained by means of combining CRISPR gene editing technology with chemogenetic activation technology, can be used for knocking out vGAT of a GABAergic neuron, and can activate the GABAergic neuron with the vGAT knocked out by means of the chemogenetics, so as to achieve the purpose of long-term activation of the GABAergic neuron with the vGAT knocked out.
