Abstract:
Provided are compounds that selectively bind to switch I adjacent groove (SWAG) site on a GTPase enzymes, such as large GTPase enzymes. For example, the GTPase enzyme can be dynamin-related protein 1 (Drp1) or other dynamin superfamily proteins. Also provided are methods of selectively inhibiting mitochondrial dysfunction by binding such a compound to the SWAG site. Provided are methods of treating a patient for mitochondrial dysfunction by administering such compounds. For instance, the patient can have mitochondrial dysfunction caused by inflammatory cell stress from sepsis from an infection, a neurodegenerative disease such as Parkinson's disease or multiple sclerosis (MS), ischemia such as a heart attack, or inflammatory disease such as inflammatory bowel disease (IBD).
