Abstract:
The disclosure technique of snail detoxification before mucus collection, process of preparation including of mucus collection by dropper spray water at snail's back (mantle), proper retention of mucus and filtration process can get potential snail mucus of higher concentrated, cleanliness and found important substances for using in skin care preparations and fractionation of glycosaminoglycans, hyaluronic acid, heparin, heparin sulfate, chondroitin by fast protein liquid chromatography (FPLC).

CLAIMS

1. Mucus collection process, start with process of snail detoxification preparation of feeding snails with unbleached brown tissue paper overnight, wet tissue by clean water then shed into small pieces and lay wet tissue 2 gram weight per snail each in their food plate. By the fact that the unbleached brown tissue as clean cellulose will cleanliness snail’s digestive system.

2. Mucus collection process according to claim 1, in accordance with the collectors wear rubber gloves and mask, prepare 10 ml clean water in container. Use one hand to hold snail’s shell and the another hand use dropper spray water at snail’s back (mantle), spraying water repeatedly until water turn to viscosity liquid and cannot suck, however this process must not spend time over 30 minutes. About 0.5 ml of mucus was obtained from each snail under 1 minute collection spend. Pour the mucus in cold resistant plastic bag and stacked in an opaque container before storage in freezer temperature -15 to -18 degree Celsius. When will be used, placed the frozen mucus at room temperature until dissolve into a viscous liquid, then filter with fine filter paper 0.25 micron for separating microorganisms and contaminants out. Until getting clear liquid then storage in condition lower 3 degree Celsius and packing in sealed container. Mucus was obtained by said process, found 100 times protein content more than the former method.

Its use mucus liquid as composition in skin care preparations or fractionation of Glycosaminoglycans (Hyaluronic acid, heparin, heparin sulfate, chondroitin) for pharmaceutical industry accordingly.

Background Art

The snail mucus is excreted from the cells in the epithelium of the skin, foot and body. For the purpose of maintain moisture, adhesive locomotion, mating and hunting, especially a thin layer of pedal mucus is secreted for crawling. Whilst, the viscous mucus are a thick layer and high concentration excreted from body skin. The major biochemical component of snail mucus is a glycosaminoglycan molecules contain sulfated mucopolysaccharide, soluble proteins, uronic acid and oligoelements.

The important role of antibiotic peptide such as mucin of the snail mucus exhibited positive antibacterial activity both for the Gram positive and Gram negative bacteria. Moreover, antimicrobial peptides are important to immune defense by acting not only as natural antibiotics but also stimulate the immune system, including barrier repair and inflammatory cell recruitment. Currently, there are many applications of snail mucus in skin care cosmetics and pharmaceutical products.

In the past still had not yet ever studied the best preparation & collection process of snail mucus same as this said invention. Due to the techniques are poorly described in records.

WO 2013/011371 relates to an apparatus for self-use in the extraction of slime from snail without any essential preparation of snails detoxification and proper mucus retention.

PAT-FR273 4725 disclose extraction of snail mucus by salt solution stimulation at a variable voltage of 50-100 V, for max.12 minutes however no any snail’s detoxification and protein value comparison between the former method with said invention.

PAT-WO2016159665A3, relates to a cosmetic composition comprising slime of snail fed with berry fruits and a preparation method thereof. The invention still not completed in comparison of mucus quality between berry fruits fed and many veggies fed. PAT-US20170281690A1, relates to a pharmaceutical composition for preventing, treating and curing rosacea comprising snail slime, chamomile and propolis. The invention is descriptive term applied to rosacea treatment although snail slime extracted can be utilized in wide band medicinal applications.

PAT-EP 3124030A1, relates to a method for producing snail extract from cornu aspersum and its extract, comprising at least two steps, being remarkable in that a first step for obtaining a non stressful snail extract acquisition and drying with a subsequent resuspension step of the produced snail extract. The invention don’t any snail’s detoxification and mucus collection process similar with said invention.

US Patent Application # 20170216368, the invention relates to a pharmaceutical for preventing, treating, and curing psoriasis including snail slime, chamomile, and honey. The invention claims a method for obtaining the composition and the use thereof to prepare a drug or device for preventing, treating or curing psoriasis lesions on the head, the skin of the body or of the face. However the invention don’t indicate the best process of snail mucus obtained.

PAT-EP3135107A2, relates to use of an ozonization device for the extraction of snail slime and related device, stimulating solution and process for the extraction of snail slime. The invention concerns a relative device, a stimulation solution and a process for extracting snail slime. However no any snail’s detoxification process and protein value comparison between the former method with said invention.

Bungon Kongin (2012) Biology of Terrestrial Snail Secretion and Application disclose the major biochemical component of snail mucus and applications without any mucus collection process.

S.J. Pancake and Manfred L. Karnovsky (1970) The Isolation and Characterization of a Mucopolysaccharide Secreted by the Snail, Otella lactea describe the mucus was obtained by holding the snails upside down and stroking the foot with a Pasteur pipette causing the snail to secrete the mucus. About 0.2 ml of mucus was obtained from each snail each week, this method may cause snail injury and get a few important substances. The present invention relates to a key process for the mucus collection relate to snail detoxification, spraying clean water on the snail back (mantle), proper retention of mucus and filtration the mucus before using meet to get potential mucus of higher concentrated with corresponding more benefits achieved in using as active ingredient in cosmetic industry and fractionation of Glycosaminoglycan (GAG) in pharmaceutical industry.

Disclosure of Invention

The newly said technique of snail mucus collection process consist of snail detoxification, spraying clean water on the snail back (mantle), proper retention of mucus and filtration the mucus before using.

Mucus was obtained by said process, found 100 times protein content more than the former method was generally using rounded glass rod pat at the snail’s body skin and foot.

The analysis of mucus’s protein content by electrophoresis comparison this newly said technique with the former method as per Fig.l

Snail back (mantle) is a suitable area closely with its shell found mucus enriched important substances when spraying clean water, 10 drops (0.5 ml) snail mucus can be obtained with this effort.

The protein content of this technique (B) has high concentration 2,530 mg/liter measured by Bradford assay, compare the former method (A) found 20-30 mg/liter only.

Snail mucus collection process as per this said technique has much more significant benefits achieved than the former method on protein components.

Process of snail detoxification

1 The yesterday night, raised snails with unbleached (brown) tissue paper, wet tissue by clean water then shred into small pieces and lay wet tissue weight 2 gram per snail each in their food plates.

2 The next morning, cleaning snails one by one without contaminants then wait in clean room temperature for collecting the mucus. Mucus Collection Process

3 The collectors wear rubber gloves and mask, prepare 10 ml clean water in container. Use one hand to hold snail’s shell and the another hand use dropper spray water at snail’s back (mantle) as per drawing (Fig.2)

4 Spraying water repeatedly until water turn to viscosity liquid and cannot suck, however this process must not spend time over 30 minutes. About 0.5 ml of mucus was obtained from each snail under 1 minute collecting spend.

5 Pour the mucus in cold resistant plastic bag and stacked in an opaque container before storage in freezer temperature -15 to -18 degree Celsius.
