The invention provides a chemical entity of Formula (I) wherein R1, R2, X, Y and Z have any of the values described herein, and compositions comprising such chemical entities; methods of making them; and their use in a wide range of methods as disclosed herein, including metabolic and reaction kinetic studies; detection and imaging techniques; radioactive treatments; modulating and treating disorders mediated by PDE2 activity; treating neurological disorders, CNS disorders, dementia, neurodegenerative diseases, and trauma-dependent losses of function; treating stroke, including cognitive and motor deficits during stroke rehabilitation; facilitating neuroprotection and neurorecovery; enhancing the efficiency of cognitive and motor training, including animal skill training protocols; and treating peripheral disorders, including hematological, cardiovascular, gastroenterological, and dermatological disorders.

AMENDED CLAIMS

What is claimed is:

1. of Formula (I) :

(I)

wherein

X is -CH2- or ·()·.

Y is -C I I - or -CF2~;

Z is -( ί ! - or -C(=0)-;

R1 is a member selected from the group consisting of:

(a) phenyl unsubstituted or substituted with one, two, three, four, or five Ra members;

where Ra is each independently selected from the group consisting of: -H, -halo, -Ci-ealkyl, -Ci-ehaloalkyl, -Ci-ealkoxy, -Ci-ehaloalkoxy, -CN, -N(Ci-6 lkyl)2, -SF5, -Cs-ecycloalkyl, -pyrrolidine, -morpholine, -piperidine, -pyrazoie, -furan, -imidazole, -thiophene, -thiazole, -pyridine, and -phenyl, wherein phenyl is unsubstituted or substituted with one, two, three, four, or five Rb members;

where R is each independently selected from the group consisting of: -H, -CI and -F; or optionally two adjacent Ra members come together to form a ring, each optionally unsubstituted or substituted with one or more members independeniy selected from: -H, -halo, -Ci-eaikyl, or -Ci-ealkoxy;

(b) monocyclic or bicyclic heteroaromatic ring each unsubstituted or substituted with one, two, three, or four members;

where R° is each independently selected from the group consisting of: -H, -halo, -Ci-ealkyl, ~C1.4haioa.lkyl, -N(Ci.6a3kyl)2, -(Ci.6alkyl)cycloalkyl, -cyclopropyl, -morpholine, -pyrrolidine, -4-chlorophenoxy, and -phenyl optionally unsubstituted or substituted with -halo, -Ci.ealkyl, or and

(c) heterocycloalkyl ring unsubstituted or substituted with one or more -H, -F, or -OCH3; and R2 is -Ci-ealkyl.

2. A chemical entity as in claim 1, wherein X is -O- and Y is -CH2~.

3. A chemical entity as in claim 1 , wherein X is ~CH2- and Y is -CF2-.

4. A chemical entity as in claim 1 , wherein X is ~CH2- and Y is -CH2-,

5. A chemical entity as in claim 1, wherein Z is -(¾-.

6. A chemical entity as in claim 1, wherein Z is -C(=0)-.

7, A chemical entity as in claim 1, wherein R is -CH3.

8. A chemical entity as in claim 1 , wherein R! is phenyl unsubstituted or substituted with one, two, three, four, or five Rd members.

9. A chemical entity as in claim 1, wherein R3 is a member independently selected from the group consisting of: -H, -halo, -Cj-ealkyl, -Ci-ehaloalkyl, -Ct-ealkoxy, -Ci-ehaloalkoxy, -CN, -N(Ci-6alkyl)2, and -C3-6cycloalkyl.

10. A chemical entity as in claim 1, wherein Ra is a member independently selected from:

-H, -CI, -Br, -F, -I, -G. alkyl, -Ci.4haloalkyl, -C5.. aikoxy, -Ci..4haioalkoxy, -CN, -N(CH3)2, and -cyclopropyl.

11 . A chemical entity as in claim 1 , wherein Ra is a member independently selected from:

-pyrrolidine, -morpholine, -piperidine, -pyrazole, -furan, -imidazole, -thiophene, -thiazole,

-pyridine, or phenyl substituted with one or more -H, -CI, or -F.

A chemical entity as in claim 1, wherein R1 is naphthalen-l -yl, naphthalen-2-yl, 6- methoxynaphthalen-2-yl, l-methoxynaphthalen-2-yl, 3-methoxynaphthalen-2-yl, 3- methoxynaphthalene-2-yl, 6-fluoronaphthalen-2-yl, 5,6,7,8-tetrahydronaphthalen-2-yl, 5,6,7,8-tetrahydronaphthalene-2-yl, 2,3-dihydro-lH-inden-5-yl, 2,3-dihydro-lH-inden-5-yl,

2H-l,3-benzodioxol-4-yl, 2H-l,3-benzodioxol-5-yl, 2,3-dihydro-l,4-benzodioxin-6-yl, 2,2- difluoro-2H-l ,3-benzodioxol-5-yl, 1 -ethyl- l H-indole, indole, isoquinoline, 2,3-dihydro-l- benzofuran-5 -yl, 2 ,3 -dihydro- 1 -benzofuran-6-yl, 3 ,4-dihydro-2H- 1 -benzopy ran-6-yl, 1 - benzofuran, l-benzofuran-5-yl, l ,3-benzothiazol-6-yl, or l-benzothiophen-5-yl.

14. A chemical entity as in claim 1, wherein R! is chroman-2-yl, 6-fluorochroman-2-yl, chroman-3-yl, 7-methoxychroman-3-yl, 2,3-dihydrobenzofuran-2-yl, (2,3- dihydrobenzo [b] [ 1 ,4] dioxin-2-yl, or 2-methyl- 1 ,2, 3 ,4-tetrahydroisoquinolin-3-yl .

15. A chemical entity as in claim 1 , wherein R1 is phenyl substituted with two or three R" members independently selected from the group consisting of: -H, -halo, and -Ci-ehaloalkyl.

A chemical entity as in claim 1, wherein Z is -C=0, -R1 is 2,3-dihydro-l-benzofuran, 3,4-dihydro-2H-l-benzopyran, benzofuran, benzothiophene, naphthalene, quinoline, or phenyl, wherein phenyl is unsubstituted or substituted with two, three, or four Ra members, where R3 is a member independently selected from: -H, -CI, -Br, -F, -I, -Chalky!, -Ci-4haloalkyi, -Ci-4alkoxy, -Ci_4haloalkoxy, -OCF2H, -CN, -N(CH3)2, or -cyclopropyl.

17. A chemical entity as in claim. 1, wherein Z is -CH2, ~R! is phenyl unsubstituted or substituted with two, three or four Ra members, where Ra is a member independently selected

23. A method of treating a neurological disorder, comprising administering to a subject in need thereof an effective amount of a chemical entity of a claim 1, 18, or 19,

wherein the neurological disorder is selected from the group consisting of a central nervous system (CNS) disorder, a developmental disorder; a schizophrenia spectrani or psychotic disorder; a depressive disorder; an anxiety disorder; an obsessive-compulsive disorder; a dissociative disorder; a disruptive, impulse-control, or conduct disorder; a

300 trauma- or stressor-related disorder; a feeding or eating disorder; a sleep-wake disorder; a sexual disorder; a substance-related or addictive disorder; and a personality disorder.

24. The method of claim 23, wherein the method is directed to a cognitive deficit due to the neurological disorder.

25. A metliod of treating a neurological disorder, comprising administering to a subject in need thereof an effective amount of a chemical entity of claim 1, 18, or 19,

wherein the neurological disorder is an acquired disorder selected from the group consisting of delirium, dementia, an age-associated cognitive deficit, a trauma-dependent loss of function, and a cognitive impairment due to chemotherapy.

26. The method of claim 25, wherein said neurological disorder is dementia due to a neurodegenerative disease.

27. The method of claim 26, wherein said neurodegenerative disease is selected from the group consisting of Alzheimer's disease, Parkinson's disease, Huntington's disease, Lewy body disease, Pick's disease, a prion disease. Amyotrophic lateral sclerosis (ALS), multiple sclerosis, frontotemporal lobar degeneration, and corticobasal degeneration.

28. The method of claim 25, wherein said age-associated cognitive deficit is age-associated memory impairment (AAMI) or Mild Cognitive Impairment (MCI).

29. The method of claim 25, wherein said trauma-dependent loss of function is selected from the group consisting of stroke, traumatic brain injury (TBI), head trauma, and head injury.

30. The method of claim 29, wherein said trauma-dependent loss of function is an impairment in motor function .

31. The method of claim 29, wherein said trauma-dependent loss of function is an impairment in cognitive function.

32. A method of treating a neurological deficit during stroke rehabilitation, comprising: a. administering to a subject in need thereof a chemical entity of any one of claims 1 , 18, or 19 during recovery of said subject from stroke;

b. training said subject under conditions sufficient to improve performance of a neurological function whose impairment is associated with said deficit;

c. repeating steps (a) and (b) one or more times, whereby the amount of training sufficient to improve said performance is reduced compared to that produced by training

33. The method of claim 32, wherein said deficit is a motor deficit.

34. Tire method of claim 32, wherein said deficit is a cognitive deficit.

35. The method of claim 34, wherein said cognitive deficit is a deficit in memory formation. 36. The method of claim 35, wherein said deficit in memory formation is a deficit in long- term memory formation.

37. The method of claim 32, wherein said training comprises a battery of tasks directed to the neurological function.

38. The method of claim 32, wherein said administering step (a) occurs in conjunction with sai d training step (b) .

39. A method of treating a disorder, comprising administering to a subject in need thereof an effective amount of a chemical entity of claim 1, 18, or 19,

wherein said disorder is selected from the group consisting of an infectious disease, a hematological disease, a cardiovascular disease, a gastroenterological disease, and a dermatological disease.

40. A method of treating a deficit during TBI rehabilitation, comprising:

a. administering to a subject in need thereof a chemical entity of any one of claims 1, 18, or 19 during recovery of said subject from TBI;

b. training said subject under conditions sufficient to improve performance of a neurological function whose impairment is due to said deficit;

c. repeating steps (a) and (b) one or more times, whereby the amount of training sufficient to improve said performance is reduced compared to that produced by training alone.

41. The method of claim 40, wherein said deficit is a motor deficit.

42. The method of claim 40, wherein said deficit is a cognitive deficit.

43. The method of claim 40, wherein said TBI is a brain trauma, a head trauma, or a head injury.

44, The method of claim 40, wherein said administering step (a) occurs in conjunction with said training step (b),

45. A method of enhancing the efficiency of an animal skill training protocol, comprising:

302 a. administering to a non-human animal a chemical entity of any one of claims 1, 18, or 19;

b. providing training to said non-human animal under conditions sufficient to improve performance of one or more skills: and

c. repeating steps (a) and (b) one or more times, whereby the amount of training sufficient to improve said performance is reduced compared to that produced by training alone.

46. The method of claim 45, wherein said animal is a sen/ice animal.

47. The method of claim 46, wherein said service animal is a dog.

48. The method of claim 45, wherein said training protocol is directed to a single skill.

49. The method of claim 45, wherein said training protocol is directed to multiple skills.

50. The method of claim 45, wherein said administering step (a) occurs in conjunction with said training step (b).
