Disclosed is a compound of formula (1), or a pharmaceutically acceptable salt thereof, where R1, R2, R3, R4, R5, R6, R7, R7', R8, R9, R10, R11, R12 and R13 are as disclosed herein. Also, disclosed is a process for the preparation of the compound of formula 1, or a pharmaceutically acceptable salt thereof, and5 intermediates used therein. The compound of formula (1) can be used in the preparation of halichondrin analogs, such as Eribulin; and a process for its preparation from the compound of formula (1) is also disclosed.

WHAT IS CLAIMED IS :

1. The compound of formula 1, or a pharmaceutically acceptable salt thereof:

wherein R1 and R2 each independently is H, a silyl group, an acyl group, a sulfonyl group or an alkoxycarbonyl group; or one of R1 and R2 is absent and the other R1 or R2 together with the nitrogen atom to which it is attached form an azide;

R3 is H or an alcohol protecting group; or R3 and one of R1 and R2 together form -C(=0)-, -C(=0)-C(=0)- or

-C(R14)(R15)-, wherein R14 and R15 each independently is H or a hydrocarbon, the hydrocarbon optionally having one or more heteroatoms;

R4 is H, Ci-3 alkyl or Ci-3 haloalkyl or an alcohol protecting group; R5 is H or -S02Ar, wherein Ar is an aryl group;

R6 is OR16, wherein R16 is H or an alcohol protecting group;

R7 and R7 together form a =0 or a protected geminal diol, or one of R7 and R7' is H and the other is a leaving group or a functional group that can be converted into a leaving group;

or R6 and one of R7 and R7' together form -0-, and the other R7 or R7' is H;

R8 is -C(=0)R17 or -CH2OR18; wherein

R17 is H or OR19, wherein R19 is H or a hydrocarbon, the hydrocarbon optionally having one or more heteroatoms;

R18 is H or an alcohol protecting group;

R9 is a halide or a sulfonate;

or R8 and R9 together form -C(=0)- or -CH(OR20)-; wherein R20 is H or an alcohol protecting group;

R10, R11 and R12 each independently is H or an alcohol protecting group; is a single or double bond; and

R13 is =0 or -OR21, wherein R21 is H or an alcohol protecting group.

2. The compound according to claim 1, or a pharmaceutically acceptable salt thereof, wherein the compound has the structure of formula la :

The compound according to claim 1 or 2, wherein R10, R11 or R12 is an alcohol protecting group.

The compound according to claim 1 or 2, wherein R10, R11 or R12 is a silyl protecting group.

The compound according claim 3, wherein R10, R11 or R12 is tert-butyldimethyl silyl (TBS).

The compound according to any one of claims 1 to 5, wherein R is =0. The compound according to any one of claims 1 to 6, wherein R9 is I.

8. The compound according to any one of claims 1 to 7, wherein R8 is -C(=0)H .

9. The compound according to any one of claims 1 to 6, wherein R8 and R9

together form -C(=0)-.

The compound of formula 2, or a pharmaceutically acceptable salt thereof:

wherein R1 and R2 each independently is H, a silyl group, an acyl group, a sulfonyl group or an alkoxycarbonyl group; or one of R1 and R2 is absent and the other R1 or R2 together with the nitrogen atom to which it is attached form an azide;

R3 is H or an alcohol protecting group; or R3 and one of R1 and R2 together form -C(=0)-, -C(=0)-C(=0)- or

-C(R14)(R15)-, wherein R14 and R15 each independently is H or a hydrocarbon, the hydrocarbon optionally having one or more heteroatoms;

R4 is H, Ci-3 alkyl or Ci-3 haloalkyl or an alcohol protecting group; one of R5' and R5" is H and the other is -CH2OR28 or -CH2S02-Ar, or R5' and R5" taken together form =CH-S02-Ar, wherein

R28 is H or an alcohol protecting group; and

Ar is an aryl group;

R6 is OR16, wherein R16 is H or an alcohol protecting group;

R7 and R7' together form a =0 or a protected geminal diol, or one of R7 and R7' is H and the other is a leaving group or a functional group that can be converted into a leaving group;

or R6 and one of R7 and R7' together form -0-, and the other R7 or R7' is H;

R8 is -C(=0)R17 or -CH2OR18; wherein

R17 is H or OR19, wherein R19 is H or a hydrocarbon, the hydrocarbon optionally having one or more heteroatoms;

R18 is H or an alcohol protecting group.

11. The compound according to claim 10, or a pharmaceutically acceptable salt thereof, wherein the compound has the structure of formula 2a :

The compound according to any one of claims 1, 2, 3, 4, 5, 6, 7, 10 and 11, wherein R8 is -CH2OR18, wherein R18 is H or an alcohol protecting group.

The compound according claim 12, wherein R is tert-butyldipheynyl (TBDPS).

The compound of formula 5, or a salt thereof:

wherein,

R1 and R2 each independently is H, a silyl group, an acyl group, a sulfonyl group or an alkoxycarbonyl group; or one of R1 and R2 is absent and the other R1 or R2 together with the nitrogen atom to which it is attached form an azide;

R3 is H or an alcohol protecting group; or R3 and one of R1 and R2 together form -C(=0)-, -C(=0)-C(=0)- or -C(R14)(R15)-, wherein R14 and R15 each independently is H or a hydrocarbon, the hydrocarbon optionally having one or more heteroatoms; R4 is H, Ci-3 alkyl, Ci-3 haloalkyl or an alcohol protecting group; one of R5' and R5" is H and the other is -CH2OR28 or -CH2S02-Ar, or R5' and R5" taken together form =0 or =CH-S02-Ar, wherein

Ar is an aryl group; and

R6' is -CH2-CH =CR29R29', -CH2C(=0)-R25 or -CH2-CH2-0-R26, wherein

R29 and R29' each independently is H or a hydrocarbon, the hydrocarbon optionally having one or more heteroatoms;

R25 is H or OR27, wherein R27 is H or a hydrocarbon, the hydrocarbon optionally having one or more heteroatoms; R26 is H or an alcohol protecting group; or R6' and one of R5' and R5" together form a protected vicinal diol.

The compound according to claim 14, or a salt thereof, wherein the

compound has the structure of formula 5a :

16. The compound according to claim 14 or 15, wherein R5' is H and R5" is

-CH2S02-Ar, wherein Ar is an aryl group.

17. The compound according to any one of claims 14 to 16, wherein R6' is

-CH2C(=0)-R25, and wherein R25 is H or OR27, wherein R27 is H or a

hydrocarbon, the hydrocarbon optionally having one or more heteroatoms.

18. The compound according to any one of claims 1 to 17, wherein R3 is H, a silyl group, an acyl group or an alkoxycarbonyl group.

19. The compound according to any one of claims 1 to 18, wherein R1 and R2 each independently is H, a silyl group, an acyl group, a sulfonyl group or an alkoxycarbonyl group, and at least one of R1 and R2 is other than H .

20. The compound according to any one of claims 1 to 17, wherein R3 and one of R1 and R2 together form -C(=0)-, and other R1 or R2 is H, a silyl group, an acyl group or an alkoxycarbonyl group.

21. The compound according to any one of claims 1 to 20, wherein R4 is Ci-3 alkyl group.

22. The compound according to any one of claims 1 to 20, wherein R4 is benzyl.

23. A process for preparation of a compound of formula 3, or a pharmaceutically acceptable salt thereof, the process comprising

performing an intramolecular cyclization reaction on a compound of formula lb to form the compound of formula 3, wherein

R1 and R2 each independently is H, a silyl group, an acyl group, a sulfonyl group or an alkoxycarbonyl group; or one of R1 and R2 is absent and the other R1 or R2 together with the nitrogen atom to which it is attached form an azide;

R3 is H or an alcohol protecting group; or R3 and one of R1 and R2 together form -C(=0)-, -C(=0)-C(=0)- or

-C(R14)(R15)-, wherein R14 and R15 each independently is H or a hydrocarbon, the hydrocarbon optionally having one or more heteroatoms; R4 is H, Ci-3 alkyl or Ci-3 haloalkyl or an alcohol protecting group;

R5 is H or -S02Ar, wherein Ar is an aryl group;

R10, R11 and R12 each independently is H or an alcohol protecting group;

is a single or double bond; and

R13 is =0 or -OR21, wherein R21 is H or an alcohol protecting group.

24. A process for preparation of a compound of formula 1, or a pharmaceutically acceptable salt thereof, the process comprising :

coupling a compound of formula 2b with a compound of formula 4 to form the compound of formula 1; wherein

R1 and R2 each independently is H, a silyl group, an acyl group, a sulfonyl group or an alkoxycarbonyl group; or one of R1 and R2 is absent and the other R1 or R2 together with the nitrogen atom to which it is attached form an azide;

R3 is H or an alcohol protecting group; or R3 and one of R1 and R2 together form -C(=0)-, -C(=0)-C(=0)- or

-C(R14)(R15)-, wherein R14 and R15 each independently is H or a hydrocarbon, the hydrocarbon optionally having one or more heteroatoms;

R4 is H, Ci-3 alkyl or Ci-3 haloalkyl or an alcohol protecting group;

R5 is H or -S02Ar, wherein Ar is an aryl group;

R6 is OR16, wherein R16 is H or an alcohol protecting group;

R7 and R7' together form a =0 or a protected geminal diol, or one of R7 and R7' is H and the other is a leaving group or a functional group that can be converted into a leaving group;

or R6 and one of R7 and R7' together form -0-, and the other R7 or R7' is H ;

R8 is -C(=0)R17 or -CH2OR18; wherein

R17 is H or OR19, wherein R19 is H or a hydrocarbon, the hydrocarbon optionally having one or more heteroatoms;

R18 is H or an alcohol protecting group;

R9 is a halide or a sulfonate; or R8 and R9 together form -C(=0)- or -CH(OR20)-; wherein R20 is H or an alcohol protecting group;

R10, R11 and R12 each independently is H or an alcohol protecting group; is a single or a double bond; R13 is =0 or -OR21, wherein R21 is H or an alcohol protecting group; and

R13' is -C(=0)R22, wherein R22 is H or OR23, wherein R23 is H or a

hydrocarbon, the hydrocarbon optionally having one or more heteroatoms.

25. The process according to claim 24, wherein the coupling reaction is

performed using a base.

26. The process according to claim 25, wherein the base is n-butyllithium .

27. A process for preparation of compound of formula 2, or a pharmaceutically acceptable salt thereof, the process comprising :

coupling a compound of formula 5b with a compound of formula 6, wherein R1 and R2 each independently is H, a silyl group, an acyl group, a sulfonyl group or an alkoxycarbonyl group; or one of R1 and R2 is absent and the other R1 or R2 together with the nitrogen atom to which it is attached form an azide;

R3 is H or an alcohol protecting group; or R3 and one of R1 and R2 together form -C(=0)-, -C(=0)-C(=0)- or

-C(R14)(R15)-, wherein R14 and R15 each independently is H or a hydrocarbon, the hydrocarbon optionally having one or more heteroatoms;

R4 is H, Ci-3 alkyl or Ci-3 haloalkyl or an alcohol protecting group; one of R5' and R5" is H and the other is -CH2OR28 or -CH2S02-Ar, or R5' and R5" taken together form =CH-S02-Ar, wherein

R28 is H or an alcohol protecting group; and

Ar is an aryl group;

R6 is OR16, wherein R16 is H or an alcohol protecting group;

R7 and R7' together form a =0 or a protected geminal diol, or one of R7 and R7' is H and the other is a leaving group or a functional group that can be converted into a leaving group;

or R6 and one of R7 and R7' together form -0-, and the other R7 or R7' is H;

R6' is -CH2C(=0)R25 or -CH2CH2OR26; wherein

R25 is H or OR27, wherein R27 is H or a hydrocarbon, the hydrocarbon optionally having one or more heteroatoms;

R26 is H or an alcohol protecting group;

R8 is -C(=0)R17 or -CH2OR18; wherein

R17 is H or OR19, wherein R19 is H or a hydrocarbon, the hydrocarbon optionally having one or more heteroatoms;

R18 is H or an alcohol protecting group; and R is a halide or a sulfonate.

28. The process according to claim 27, wherein R6' is -CH2C(=0)H, and the

coupling reaction is performed using a nickel/chromium catalyst.

29. A process for preparation of the compound of formula 5, or a salt thereof, the process comprising :

- converting the terminal alcohol of the compound of formula 7 into an amine or substituted amine to form the compound of formula 5

wherein

R1 and R2 each independently is H, a silyl group, an acyl group, a sulfonyl group or an alkoxycarbonyl group; or one of R1 and R2 is absent and the other R1 or R2 together with the nitrogen atom to which it is attached form an azide;

R3 is H or an alcohol protecting group; or R3 and one of R1 and R2 together form -C(=0)-, -C(=0)-C(=0)- or - C(R14)(R15)-, wherein R14 and R15 each independently is H or a hydrocarbon, the hydrocarbon optionally having one or more heteroatoms;

R4 is H, Ci-3 alkyl or Ci-3 haloalkyl or an alcohol protecting group; one of R5' and R5" is H and the other is -CH2OR28 or -CH2S02-Ar, or R5' and R5" taken together form =0 or =CH-S02-Ar, wherein

R28 is H or an alcohol protecting group; and

Ar is an aryl group; and

R6' is -CH2-CH =CR29R29', -CH2C(=0)-R25 or -CH2-CH2-0-R26, wherein R29 and R29' each independently is H or a hydrocarbon, the

hydrocarbon optionally having one or more heteroatoms;

R25 is H or OR27, wherein R27 is H or a hydrocarbon, the hydrocarbon optionally having one or more heteroatoms; and

R26 is H or an alcohol protecting group.

30. A process for preparation of Eribulin, comprising the process as defined in any one of claims 23 to 29.

31. A process for preparation an analog of halichondrin, comprising the process as defined in any one of claims 23 to 29.

32. Eribulin mesylate having a purity of more than 95%, 96%, 97%, 98% or 99%, as determined byHPLC.

33. /V-(tert-butoxycarbonyl) eribulin

3a wherein,

R1 is an acyl group, a sulfonyl group or an alkoxycarbonyl group;

R2 and R3 are both H or together form -C(=0)-, -C(=0)-C(=0)- or

35. The compound of claim 34, wherein R1 is tert-butoxycarbonyl, and R2 and R3 together form -C(=0)-. The compound of claim 34, wherein R1 is tert-butoxycarbonyl, and R2 and R3 together form -C(Me)2-.

SYNTHETIC PROCESS FOR PREPARATION OF MACROCYCLIC Cl-KETO ANALOGS OF HALICHONDRIN B AND INTERMEDIATES

USEFUL THEREIN

CROSS REFERENCE TO RELATED APPLICATIONS

[0001] This application claims the benefit of and priority to US Provisional Application Numbers 61/618,004 filed March 30, 2012, entitled MACROCYCLIC ANALOGS AND METHODS FOR THEIR PREPARATION, and 61/647,127 filed May 15, 2012, entitled SYNTHETIC PROCESS FOR PREPARATION OF MACROCYCLIC Cl- KETO ANALOGS OF HALICHONDRIN B AN D INTERMEDIATES USEFUL THEREIN . The contents of the above applications are incorporated herein by reference.

FIELD

[0002] The specification relates to a synthetic process for preparation of macrocyclic Cl-keto analogs of Halichondrin B, their salts and intermediates useful for their preparation.

BACKGROUND

[0003] Halichondrins have been disclosed as having anti-cancer and

antimitotic activity (Chem. Rev. 2009, 109, 3044-3079, incorporated herein by reference). In particular, Halichondrin B has been reported as a potent anticancer agent that was first isolated from the marine sponge Halichondria okadai (US 5,436,238; Tetrahedron Lett. 1994, 35, 9435 and WO 1993/017690 Al, all incorporated herein by reference). It was further reported that analogs of

Halichondrin B bearing only macrocyclic fragment of its molecule (CI - C30 fragment) and having a ketone function instead of ester at CI position demonstrate anticancer activity similar to Halichondrin B (Bioorg. Med. Chem . Lett., 2000, 10, 1029 and Bioorg. Med .Chem. Lett., 2004, 14, 5551) It was established that such macrocyclic fragment is responsible for induction of mitotic blocks in cancer cells via disruption of tubulin polymerization process that triggers apoptosis of cancerous cells and stops their proliferation (Cancer Res., 2004, 64, 5760 and Mol. Cane. Ther., 2008, 7, 2003). Eribulin mesylate, a macrocyclic Cl-keto analog of

Halichondrin B, has been reported as having potent anticancer properties (WO 1999/065894 Al, incorporated herein by reference). Eribulin is marketed under the trade name Halaven, and it is also known as E7389, B1939 and ER-086526.

[0004] The synthetic approach described (US 6,214,865; WO 2009/124237 Al, Bioorg. Med .Chem. Lett, 2004, 14, 5551 and J. Am. Chem. Soc. 2009, 131, 15642 , all incorporated herein by reference) involves introduction of nitrogen in the C27-C35 fragment of Eribulin after assembly of the macrocycle. Such an approach can add synthetic steps to the later stages of the synthesis, after the building blocks corresponding to the C1-C13 and C14-C26 fragments have been introduced. The synthesis of those fragments is long and complex; and every additional step in the synthesis can imply an increase in manufacturing costs. In addition, due to the cytotoxic nature of Eribulin, late introduction of the nitrogen results in a greater number of steps that can require special safety containment, which can limit throughput and can also increase the cost of producing the active pharmaceutical ingredient (API). [0005] There is a need in the art for a compound that can be used in process for preparation of Eribulin and other macrocyclic Cl-keto analogs of Halichondrin B and their salts. In addition, there is a need in the art for a compound and a process that can help to improve the convergence of the synthetic route for preparation of Eribulin and other macrocyclic CI- keto analogs of Halichondrin B and their salts, and therefore, can also help to reduce the amount of C1-C13 and C14-C26 fragments required. Further, there is a need in the art for a compound that can help to reduce the number of steps that can require safety containment for preparation of Halichondrin and its analogs. Moreover, there is a need in the art for a process for preparation of such a compound.

SUMMARY OF THE INVENTION

DESCRIPTION

[0011] As described above, in one aspect the specification relates to a compound of formula 1, or a pharmaceutically acceptable salt thereof:

A 100 mL round-bottom flask equipped with a stir bar was oven dried and cooled under inert atmosphere during introduction to the glovebox. To a 100 mL round- bottom flask equipped with a stir bar, under inert atmosphere, were added CrCI 2 (655 mg, 5.33 mmol, 1.0 eq), sulfonamide ligand Y(1.83 g, 5.86 mmol, 1.1 eq) and proton sponge (1.26 g, 5.86 mmol, 1.1 eq). The solid reagents were

suspended in acetonitrile (25 mL), producing a dark blue-green solution, which was stirred vigorously at room temperature for 1 hour. To the dark blue-green solution, lithium chloride (452 mg, 10.66 mmol, 2.0 eq), manganese (586 mg, 10.66 mmol, 2.0 eq), Zr(Cp) 2 CI 2 (1.71 g, 5.86 mmol, 1.1 eq) and a complex of nickel(II) chloride with 2,9-dimethyl-l,10-phenanthroline (NiCI 2 -dmp) (36 mg, 0.11 mmol, 0.02 eq) were added, followed by a suspension of 6a (3.47 g, 5.33 mmol, 1.0 eq) and 5j (4.09 g, 7.99 mmol, 1.5 eq) in acetonitrile (25 mL). The reaction vessel was capped and stirred vigorously for 16 hours. Another portion of lithium chloride (452 mg, 10.66 mmol, 2.0 eq) and NiCI 2 -dmp (36 mg, 0.11 mmol, 0.02 eq) were added and the reaction was left stirring at room temperature for 4 hours more. The reaction vessel was sealed with a rubber septum and brought outside the glovebox. Anhydrous THF (50 ml_) was added to the round bottom flask and the resulting mixture was stirred at room temperature for 30 minutes.

The layers were separated and the organic phase was washed with additional water (2x 10 m L), brine (10 m L), dried over MgS0 4 and concentrated under reduced pressure. The residue was dissolved in MeOH (10 m L), a catalytic amount of K 2 C0 3 was added at room temperature and the resulting mixture was stirred for 2 hours. The reaction was diluted with dichloromethane and quenched with water (10 mL). The layers were separated and the aqueous phase was further extracted with dichloromethane (5 x 10 m L). The combined organic layers were washed with brine (20 m L), dried over MgS0 4 , filtered and concentrated. A 50 mL round-bottom flask equipped with a stir bar was oven dried and cooled under inert atmosphere during introduction to the glovebox. To a 50 mL round-bottom flask equipped with a stir bar, under inert atmosphere, were added CrCI 2 (296 mg, 2.41 mmol, 1.0 eq), sulfonamide ligand (829 mg, 2.65 mmol, 1.1 eq) and proton sponge (568 mg, 2.65 mmol, 1.1 eq). The solid reagents were suspended in acetonitrile (11 mL), producing a dark blue-green solution, which was stirred vigorously at room temperature for 1 hour. To the dark blue-green solution, lithium chloride (204 mg, 4.82 mmol, 2.0 eq), manganese (265 mg, 4.82 mmol, 2.0 eq), Zr(Cp) 2 CI 2 (775 mg, 2.65 mmol, 1.1 eq) and NiCI 2 -dmp (16 mg, 0.05 mmol, 0.02 eq) were added, followed by a suspension of 6a (1.57 g, 2.41 mmol, 1.0 eq) and 1-23 (1.44 g, 3.62 mmol, 1.5 eq) in acetonitrile (11 m l_) . The reaction vessel was capped and stirred vigorously for 1-2 hours. Another portion of lithium chloride (204 mg, 4.82 mmol, 2.0 eq) and NiCI 2 -dmp (16 mg, 0.05 mmol, 0.02 eq) were added and the reaction was left stirring at room temperature for 16 hours. The reaction mixture was then poured into a suspension of florisil (< 200 mesh, 30 g) in ethyl acetate (200 ml_) and stirred at room temperature (rt) for 1 hour. The mixture was filtered over silica gel and rinsed several times with ethyl acetate. The filtrate was concentrated and subsequently purified by column chromatography on silica gel using 0 - 30% acetone in dichloromethane as eluent. The product E-1B was afforded as a grey foam (970 mg, 43%) and used as is.

concentration. The crude product was purified by column chromatography on silica gel using 10 - 20% acetone in dichloromethane to afford the product E-3A as a grey foam (693 mg, 74%).

A solution of TMSCI (1M in THF, 0.85 mL, 0.85 mmol, 2.2 eq) was added at 0 °C and the solution was stirred for another 10 min. A second portion of LDA (0.5M in THF, 0.92 mL, 0.46 mmol, 1.2 eq) was added and the reaction mixture was stirred for another 10 min at 0 °C. Compound 4a (394 mg, 0.53 mmol, 1.4 eq) was dissolved in THF (3 mL) and added to the reaction mixture, which was stirred at 0 °C for an additional 30 min. The reaction was quenched with the addition of saturated aqueous ammonium chloride solution (10 mL) and the resulting mixture was extracted with MTBE (3x 10 mL). The combined organic layers were washed with brine (20 mL), dried over MgS0 4 , filtered and concentrated. The crude material was separated into two lots (E-5A and E-5Ab) by column chromatography on silica gel using 10 - 30% acetone in

dichloromethane. E-5A was obtained as a white foam (150 mg, 28%).

The reaction vessel was sealed with a rubber septum and brought outside the glovebox. Anhydrous THF (5 mL) was added to the round bottom flask and the resulting mixture was stirred at room temperature for 30 minutes.
