Abstract:
Provided are a T lymphocyte modified by double chimeric antigen receptor and a preparation method thereof. A chimeric antigen receptor (CAR-L) with a low affinity and a chimeric antigen receptor with a high affinity (CAR-H) are constructed, and are used for respectively recognizing two tumor-associated antigens, and respectively contain a CD3ζ sequence and a costimulatory molecule (costimulatory molecule, CM) signal sequence. The CAR-L and the CAR-H are simultaneously transfected into the T lymphocyte, and the modified T lymphocyte can be effectively activated only by simultaneously recognizing the two tumor-associated antigens, thus reinforcing the targeting of the CAR-T lymphocyte in killing tumors and reducing the damage to normal tissues.
