Abstract:
Provided is a therapeutic agent for neurological disorder, said therapeutic agent directly acting on vascular barrier failure that depends on oxygen concentration. The therapeutic agent for neurological disorder comprises, as an active ingredient, any of: (a) a functional nucleic acid to ADAM (a disintegrin and metalloproteinase) 12 or ADAM17; (b) a vector expressing the functional nucleic acid (a), said vector containing a DNA that encodes the functional nucleic acid; and (c) a neutralizing antibody to ADAM12 or ADAM17. The functional nucleic acid is preferably an siRNA. The functional nucleic acid to ADAM12 is preferably an siRNA that comprises a sense strand sequence of a nucleotide represented by SEQ ID NO:1 or 3 and an antisense strand sequence complementary thereto. Alternatively, the functional nucleic acid to ADAM17 is preferably an siRNA that comprises a sense strand sequence of a nucleotide represented by SEQ ID NO:5 or 7 and an antisense strand sequence complementary thereto.
