Abstract:
A Tripterygium wilfordii triterpene synthase TwOSC1 protein and a coding gene thereof. cDNA of a TwOSC1 gene is cloned to eukaryotic expression vector pYES2, and a recombinant expression vector carrying the TwOSC1 gene is constructed and transformed into a yeast expression host, allowing triterpenoids such as friedelin and amyrins to be produced. Mutation research has shown that amino acids 486 and 502 in the code of the TwOSC1 gene are key sites able to increase or decrease production of friedelin or amyrins. Gene gun-mediated interference experimentation has shown that interference of the TwOSC1 gene has a marked inhibiting effect on the synthesis of celastrol in Tripterygium wilfordii, and further gene gun-mediated overexpression experimentation has shown that overexpression of the TwOSC1 gene can increase production of celastrol in Tripterygium wilfordii suspension cells. TwOSC1 protein and the coding gene therefor may be used in biosynthesis of plant triterpenoids, and in cultivating high-quality Tripterygium wilfordii.
