Abstract:
The present disclosure provides use of CDK16 as a target in the preparation of a medicine for treating triple-negative breast cancer (TNBC). According to the present disclosure, research has found that the knock-down of the CDK16 can significantly inhibit the growth of TNBC xenograft tumors, PDX and PDO, and the mechanism is mainly the inhibition of phosphorylation of PRC1 (T481) and prevention of spindle apparatus formation during cell mitosis by means of the knock-down of the CDK16, which prevents the cells from dividing normally and thus inhibits the occurrence, development and metastasis of the TNBC tumors. Additionally, research has found that the covalent small-molecule inhibitor Rebastinib of the CDK16 shows a good effect in inhibiting the development of TNBC tumors, and is a potential novel medicine for treating the TNBC.
