Abstract:
Provided is the use of a polymer in the detection and preparation of cells expressing CAR. Antigen polymers can achieve the highly specific, sensitive and precise detection of CAR, can be used for sensitive detection of rare CAR-T cells by means of magnetic enrichment, and can also be used for activating phenotypes and specific proliferation by means of temperature control and specific CAR-T cell stimulation and achieving multi-dimensional analysis on CAR-T cells by means of single-cell multiomics analysis. In addition, a method for specifically producing CAR-T cells using antigen polymers is different from the current method for non-specifically activating and proliferating T cells using nonspecific anti-CD28/CD3 magnetic beads. Since the antigen-polymer only specifically activates and proliferates corresponding CAR-T cells and does not activate other T cells, there is no need to worry about the potential toxicity of magnetic beads, which is a better method for preparing CAR-T cells. Further provided is an antigen polymer labeled with an oligonucleotide, which achieves the detection of the expression of CAR by means of single cell sequencing.
