Abstract:

PURPOSE: To improve therapy and diagnosis of human cancers by bringing to contain a specific sequence having specificity to the antigen bound by A10 murine monoclonal antibody (mAb).

CONSTITUTION: A chimeric mouse-humane A10 antibody having specificity to the human tumor cell antigen is produced by obtaining a messenger RNA (mRNA) (a) by separating hybridoma cell lines of mAb-producing murine B cells, treating a complete length-cDNA library obtained by treating the component (a) to obtain DNA (b) in a variable (V) region containing light (L) chains and heavy (H) chains of immunoglobulin, while cloning the cDNA obtained from the component (a) to obtain a gene part module (c) of a constant (C) region, then, bonding the component (b) to the component (c) to obtain a plasmid pING2254 (d)-coding for the H chain or the L chain, succeedingly culturing a host obtained by introducing the component (d) into a desired procaryote and extracting the culture solution.
